松鼠猴源沙门菌的分离鉴定及生物学特性研究

摘    要:从扬州某动物园病死松鼠猴的病料中分离出疑似沙门菌株,经革兰氏染色镜检、生化试验、PCR扩增沙门菌invA基因和血清型鉴定,再对该菌株的生物学特点如致病性、生物被膜形成能力和药物敏感性进行研究。结果表明,从送检猴肝中分离出的菌株,革兰氏染色为阴性短杆菌;能够发酵葡萄糖、麦芽糖、甘露醇等,不能发酵乳糖和蔗糖,V-P反应阴性,MR阳性;PCR检测inv A基因为阳性,确认分离菌为沙门菌;血清型鉴定为鼠伤寒沙门菌。致病性试验采用灌胃方式攻毒小鼠,18 h后小鼠死亡。生物被膜检测结果表明,该分离菌株有较强的生物被膜形成能力。药敏结果显示,分离菌对头孢噻肟、头孢哌酮和卡那霉素等敏感,对庆大霉素、链霉素和复方新诺明耐药。

Abstract:A suspected Salmonella strain was isolated from the diseased material of a dead squirrel monkey in a zoo in Yangzhou.This isolate was carried on Gram staining,biochemical test,PCR amplification of Salmonella inv A gene,serotype identification,and biological characteristics such as pathogenicity,biofilm formation and drug sensitivity.The results showed that the isolate from liver of the monkeys was Gram-stained negative bacillus,could ferment glucose,maltose and mannitol,and not ferment lactose and sucrose.It was negative to VP,and positive to MR.Salmonella was identified by PCR using invA.The serotype was identified as Salmonella typhimurium.To test the pathogenicity,mice were orally administered with the isolate.Infected mice died 18 hours later.Biofilm formation assay showed that this strain could form strong biofilm.The result of the drug sensitivity results showed that this isolate was sensitive to cefotaxime,cefoperazone,and kanamycin,and resistant to gentamicin,streptomycin,and compound sulfamethoxazole.
