Amelogenin locus typing using toehold-assisted fluorescent DNA melting analysis

Abstract:The amelogenin gene is the locus of choice for gender identification in forensic science. Here we report on the use of fluorescent DNA melting curve analysis to genotype the amelogenin locus by means of a toehold-assisted DNA strand displacement reaction. The shape of the curves, or “polarity” of the melting peaks, allowed for visual discrimination between male and female DNA samples.
