Characterization of 26 miniSTR loci for improved analysis of degraded DNA samples

An additional 20 novel mini-short tandem repeat (miniSTR) loci have been developed and characterized beyond the six previously developed by our laboratory for a total of 26 non-CODIS miniSTR markers. These new markers produce short PCR products in the target range of 50-150 base pairs (bp) by moving the primer sequences as close as possible-often directly next to the identified repeat region. These candidate loci were initially screened based on their small amplicon sizes and locations on chromosomes currently unoccupied by the 13 CODIS STR loci or at least 50 Mb away from them on the same chromosome. They were sequenced and evaluated across more than 600 samples, and their population statistics were determined. The heterozygosities of the new loci were compared with those of the 13 CODIS loci and all were found to be comparable. Only five of the new loci had lower values than the CODIS loci; however, all of these were much smaller in size. This data suggests that these 26 miniSTR loci will serve as useful complements to the CODIS loci to aid in the forensic analysis of degraded DNA, as well as missing persons work and parentage testing with limited next-of-kin reference samples.

In Wirklichkeit ?hnelt aber die grundbücherliche Eintragung eines Bestandrechts eher einer Anmerkung; ihre Rechtsfolgen beschr?nken sich – und zwar blo? im Verh?ltnis zwischen Bestandgeber und Bestandnehmer ohne allgemeine dingliche Wirkung gegenüber Dritten! – auf § 1120 und § 1121 ABGB.     

The Ratio of 6β‐Hydroxycortisol to Cortisol in Urine as a Measure of Cytochrome P450 3A Activity in Postmortem Cases

Poisoning can occur with chronic accumulation of a drug due to reduced metabolic capacity; conversely, under‐treatment may occur due to an increased metabolic rate. Over half of all drugs are metabolized by the cytochrome P450 3A complex (CYP3A). The activity of CYP3A can be assessed by the urinary ratio of 6β‐hydroxycortisol to cortisol. The aim of this study was to determine the usefulness of this ratio as a postmortem marker for determining whether altered CYP3A enzyme activity occurred prior to death. In a series of 244 postmortem cases, this ratio ranged from 0.014 to 78.6 (median 3.50). The median was significantly higher (5.14) in a subgroup of 28 cases that exhibited the presence of CYP3A‐inducing drugs. In cirrhosis, the median ratio was 1.69. This pointed to a reduced metabolic capacity of CYP3A. Thus, the ratio may constitute a rough indicator of the CYP3A metabolic capacity, which could be of value in special cases.     

GC/MS衍生化法检测血中4-溴-2,5-二甲氧基苯乙胺

目的建立人体全血中4-溴-2,5-二甲氧基苯乙胺的衍生化GC/MS分析方法。方法血液样品经去离子水稀释4倍,8000r/min高速离心20min后,加入Na H2PO4缓冲溶液(p H值为6.0)2m L,混匀。将上述混合液加入用甲醇、Na H2PO4缓冲溶液(p H值为6.0)活化的Bond Elut Centify?固相萃取柱后,依次采用1.0M乙酸溶液、去离子水、甲醇、二氯甲烷/异丙醇/氨水(78/20/2,V/V/V)混合洗脱液进行提取、分离、净化处理,40℃空气流下挥干,三氟乙酸酐衍生化,GC/MS检测4-溴-2,5-二甲氧基苯乙胺和4-苯基丁胺衍生物。采用标准品衍生化产物的标准质谱图定性分析,选择m/z 242(4-溴-2,5-二甲氧基苯乙胺)、m/z 91(4-苯基丁胺)作为定量离子进行定量分析。结果血液中4-溴-2,5-二甲氧基苯乙胺的最低检出限为6ng/m L,在0.02~10μg/m L浓度范围内,线性关系良好(=0.9993),日内精密度和日间精密度均小于10%,平均提取回收率约为69%。结论该方法操作简便、灵敏度高,适用于全血中4-溴-2,5-二甲氧基苯乙胺检测。

New Panthers,old Panthers and the politics of black nationalism in the United States

ABSTRACT

The New Black Panther Party for Self-Defense has been a cause du scandale since it was formed in the early 1990s. Indeed, the Anti-Defamation League has described it as ‘the largest organized anti-Semitic and racist black militant group in America’. It is somewhat surprising, then, that there has been very little detailed analysis of the party and its activities. Mulloy begins to fill this gap by outlining the origins and development of the party, and by closely examining the ongoing dispute between the New Black Panthers and surviving members of the original Black Panther Party—established by Huey Newton and Bobby Seale in Oakland, California in 1966—over the right to claim the name, the history and the legacy of the Panthers in the United States. Critically assessing the strategies and actions of the New Black Panthers, Mulloy argues that its high-profile, media-centred approach to political activism has largely been a failure with regard to its overall aim of reviving the Black Power movement in the United States.     

云南省11个民族腺苷脱氨酶(ADA)频率的分布调查

采用电泳技术对云南省汉、彝、自、傣、哈尼、瑶、布朗、基诺、拉祜、佤及苦聪等11个民族的ADA 进行了表型分布调查。并对-20℃、4℃、室温(20℃左右)保存的血痕ADA 活性进行了测定。在汉族人群中发现了一例ADA4-1稀有型,家系调查表明其是由父亲遗传下来的。
