Species-specific differences in a non-polymorphic region of the mitochondrial cytochrome b gene appear to be large enough to allow human-specific amplification of forensic DNA samples. We therefore developed a PCR-based method using newly designed primers to amplify a 157-bp portion of the human mitochondrial cytochrome b gene. The forward and reverse primers were designed to hybridize to regions of the human mitochondrial cytochrome b gene with sequences differing from those of chimpanzee by 26% (7 bp/27 bp) and 26% (6 bp/23 bp), respectively. Using this primer pair, we successfully amplified DNA extracted from blood samples of 48 healthy adults. All these human samples produced a single band of the expected size on agarose gel electrophoresis, and the sequence of the single band was shown to be identical to that of the target region (157 bp) by sequence analysis. On the other hand, no visible bands were amplified from DNA extracted from blood samples of animals including non-human primates (chimpanzee, gorilla, Japanese monkey, crab-eating monkey) and other species (cow, pig, dog, goat, rat, chicken and tuna). Thus, DNA producing a single band following PCR amplification using this primer pair can be reasonably interpreted as being of human origin. In addition, aged biological specimens comprising bloodstains, hair shafts and bones were successfully identified as being of human origin, illustrating the applicability of the present method to forensic specimens.  相似文献

This study analyzes the bill cosponsorship behavior of reserved seat legislators in the Pakistan National Assembly, where special legislative seats are reserved for women and religious minorities. It offers a critical case because two theories on reserved seat legislators—the theory of electoral incentives and the theory of institutional weakness—lead to opposite predictions about the extent to which such legislators will engage in bill cosponsorship, with the former (latter) predicting the negative (positive) effect of reserved seats. I find that women and minorities in reserved seats initiate a greater number of bills and cosponsor bills with a larger number of peers than nonreserved male legislators. Using network analysis, I also show that reserved legislators tend to play a role in bridging different partisan blocs of legislators within the cosponsorship network. These empirical patterns provide support to the theory of institutional weakness, rather than to the theory of electoral incentives.  相似文献

A suicide caused by ingestion of multiple psychoactive drugs is reported. A 42-year-old man with a history of psychosis was found dead in a blood pool in his room. The forensic autopsy revealed two stab wounds on his chest. However, these wounds could not explain the cause of death. Eighty-six tablets were found in his stomach. Four psychoactive drugs; clocapramine (CC), chlorpromazine (CP), promethazine (PM) and clotiazepam (CT) were detected in blood and tissues. The concentrations of CC, CP, PM and CT in the femoral vein (FV) blood were 0.39, 0.61, 1.23 and 0.09 microg/ml, respectively. The cause and manner of death were attributed to suicidal multiple psychoactive drug poisoning.Postmortem drug redistribution showed great site-dependent variations with the lowest level in the FV blood. Remarkable variations were observed in CC, CP and PM, but not in CT compared to other three drugs. The variations were dependent on the volume of distribution (Vd) of the drugs. Our human case has demonstrated drugs with higher Vd values showed higher degree of postmortem redistribution of the drug and vice versa.  相似文献

A homicidal case by an air gun involving a 73-year-old woman who was found dead in her home is described. At autopsy, two gunshot wounds were observed on the left anterior chest and the right back. There was neither exit wound nor powder burns. The fatal pellet, 0.9 g and 5.5mm diameter, penetrated the left anterior chest and the heart to cause cardiac tamponade (200 ml) before lodging in the left subclavian artery. Death was due to cardiac tamponade.This case has exhibited two rare phenomena: homicide by an air gun and pellet embolisation. This is the first homicide case by use of air gun in Japan and demonstrates that although it may be rare, air guns can be used as potential homicidal weapons. This experience will alert our society and others on the potential hazards of air guns we have tended to regard as toys.  相似文献

The Orchestra of Treaties scenario emerges if countries share the following principles for re-building the climate regime (1) recognize the sovereignty concerns attending energy policies; (2) build upon national interests in technology and development; (3) avoid conflicts and enhance cooperation by appropriately framing core issues; and (4) address not only short-term emission cuts but also long-term technological change.In this scenario, countries will share these principles by drawing on the hard lessons of past UNFCCC negotiations. The framing of the climate change problem as the allocation of emission quota created an adversarial style of negotiation that resulted in distrust among countries. Moreover, the negotiations have been characterized by unpredictable outcomes. Since energy policies were deemed as sovereignty concerns, countries did not want to put such issues on the agenda to avoid a potentially intrusive outcome. This negotiation style brought about shortcomings of the Kyoto Protocol and may lead to a stalemate in future negotiations.The scenario captures the dynamics that emerge when multiple efforts are pursued by flexible coordination of actors motivated through diverse incentives. The emerging regime, the Orchestra of Treaties, will consist of four building blocks, of which three grow outside of UNFCCC.
(1)  Group of Emission Markets (GEM) begins with separate domestic markets without internationally imposed emission targets. The markets are then gradually coordinated through price signals. The advantage of this pathway is that it fosters the establishment of emission markets without conflicting with sovereignty concerns for energy policy, thereby enabling key large emitters to establish emission markets.
(2)  Zero Emission Technology Treaty (ZETT) that addresses long-term technological change. This will set zero CO2 emission from the energy sector as the long-term goal, thereby creating strong signals to stakeholders. It will begin as a non-binding pledge and review system so that it does not conflict with sovereignty concerns.
(3)  Climate-wise Development Treaty (CDT) that addresses the concerns of developing countries, which are development, adaptation, technological transfer and mitigation. In this treaty, developed countries agree to revise their assistance policies to make development more sustainable and climate-wise.
(4)  UNFCCC will serve as an information exchange arena, target funding mechanism and a political focal point.
  If the political interests and views remain diverse across countries, the Orchestra of Treaties may be the most environmentally effective regime among the alternatives. As for the evolution over time, once the technological and political feasibility of some climate policies have been demonstrated by key developed countries, all countries will be confident enough to deepen their commitments in the wider issue area. They might eventually return to a Kyoto-type structure with full participation once confidence has been built, but this may take decades.
  Concerns frequently voiced about this scenario include that the regime may impose additional negotiating burden upon developing countries, or that a departure from a Kyoto style framework may end up with losing political momentum.
