新城疫病毒F48E9株M NP F和HN基因的真核表达

将新城疫病毒(NDV)F48E9株的M、NP、F和HN基因克隆到真核细胞表达载体 pCAGG上,经酶切、PCR鉴定和序列分析,分别筛选出了含有M、NP、F和HN基因的重组质粒, 命名为pCAGG-M、pCAGG-NP、pCAGG-F和pCAGG-HN。纯化后的重组质粒通过脂质体单独转染HeLa细胞,经间接免疫荧光试验检测到了M、NP、F和HN蛋白的表达。pCAGG-M、 pCAGG-NP、pCAGG-F和pCAGG-HN重组质粒组合共转染HeLa细胞48 h后,可以观察到明显的细胞融合现象。试验结果表明,NDV F48E9株的M、NP、F和HN蛋白均在HeLa细胞内得到了成功表达,同时证明在该表达系统中F蛋白不足以诱导细胞融合。     

B. F. Skinner's utopian vision: behind and beyond walden two

This paper addresses B. F. Skinner's utopian vision for enhancing social justice and human well‐being as it was introduced in his 1948 novel, Walden Two. In the first part, we address the historical, intellectual, and social context that situates the book in the utopian genre, the critiques of its premises and practices, and the fate of intentional communities patterned on the book. In the second part, we review practices in Skinner's book that advance social justice and human well‐being under the themes of health, wealth, and wisdom, and then focus on contemporary practices that are the legacy of his vision. His vision was neither the a priori truth of a particular premise about human behavior or a necessary practice (blueprint) for an intentional community but rather the use of empirical methods to discover premises and practices that work to advance the health, wealth, and wisdom of individuals and survival of the culture.     

Brandom on Norms and Objectivity

The aim of this paper is to investigate Brandom’s conception of the objectivity of norms. In Making It Explicit Brandom supports a weak notion of objectivity based on his understanding of the perspectival structure of linguistic practices. In his following works, he resorts to the Hegelian notion of recognition, adding a historical dimension to his account. I contend that this notion of objectivity can be successfully defended against the objections raised by the commentators. In particular, it does not jeopardise the same possibility of communication, as claimed by Habermas and others, unless a strongly objective notion of communication is assumed. However, the paradigm shift from a strong to a weak understanding of objectivity entails a consequent revision of the conception of social criticism.     

EXCEL多元回归分析在痕迹数据处理上的应用

在痕迹检验技术中,很多推算公式是由回归分析预测法得出的。多元回归分析预测法的计算,虽然可以用常用计算器进行统计,但是数据复杂繁琐,工作量较大。Excel是一种办公系统软件,运用它的强大数据分析功能,只输入一次原始数据,而后全部以鼠标操作即可自动生成回归方程。文章以一组足迹数据分析为例,首先介绍相关分析,然后建立回归模型,最后计算出相关的回归方程。     

鹅副黏病毒QY分离株F基因和HN基因的克隆与序列分析

根据已发表的鹅副黏病毒基因组序列,设计并合成了扩增F基因和HN基因的5对引物,利用RT-PCR的特异性扩增出了广东省清远分离株(QY株)的F基因和HN基因。然后将其克隆入pMD 18-T载体,经鉴定、测序及拼接,QY株的F基因和HN基因全序列长度分别为1 662bp和1 716 bp,分别编码553个和571个氨基酸。经与GenBank登录的几株参考毒株F基因和HN基因编码区的核苷酸序列进行比较;结果,QY株与参考毒株SF02株和LaSota株F基因的核苷酸序列同源性分别为98.3%和82.4%,HN基因的核苷酸序列同源性分别为97.2%和75.9%。     

用乙醇和正丁醇判断豚鼠溺死时间的实验探索

本文以豚鼠为实验动物，溺死后浸泡于水温为２０℃的水缸小。运用气相色谱法测定其溺死后不同浸泡时间在心、肺、肝小＃在的乙醇和正了醇含量。实验结果表明在溺死后８天内，样本中乙醇和正了醇量随浸泡时间的延长而增加，且呈高度的直线相关。其血、肺、矸中乙醇量与溺死时间的回归方程依次为ｙ１＝－１６．８２＋１２．９５Ｘ，ｙ２＝－５８．５３＋１９．２３Ｘ，ｙ３＝－３１．９１＋４７．２９Ｘ。血、肺、肝小正丁醇量与溺死时间的回归方程依次为ｙ４＝－１０．１３＋５．８６ｘ，ｙ５＝－２９．４５＋１６．１７ｘ，ｙ６＝３．７５＋１４．３１ｘ。综合分析溺死后血、肺、肝小乙醇和正丁醇，有可能作为判断溺死时间的新指标。     

东北地区新城疫流行株的分子生物学特性调查

从东北地区发生新城疫 (ND)的病鸡、病鸽和产蛋下降鸡分离到 19株NDV ,对其融合蛋白 (F)基因 5 32bp或 2 80bp片段进行了克隆、测序 (在GenBank中的登录号为AY2 0 86 80～AY2 0 86 98) ,并对各株的F基因序列进行了比较。结果 ,各毒株之间的核苷酸同源性为 83.1%～10 0 % ,氨基酸同源性为 85 .5 %～ 10 0 % ;系统发育进化树分析表明 ,其中 2株与疫苗株V4同属基因Ⅰ型 ,为弱毒株 ;其余 17株为强毒株 ,其中 2株为基因Ⅵ型 ,其余为基因Ⅶ型。表明东北地区目前由 3种基因型的NDV毒株 (Ⅰ型、Ⅵ型、Ⅶ型 )所引发 ,并以基因Ⅶ型为主。另外 ,对其中的代表毒株APMV1/chicken/China/JL 11/ 0 2进行了免疫学试验 ,结果显示 ,LaSota疫苗的免疫保护效果不理想 ,可能是LaSota疫苗免疫鸡群发生ND的主要原因。     

EXCEL在交通事故分析中的应用

交通安全问题已引起了世界各国的重视,每个国家都在寻求和探索降低交通事故发生率的良策。我国是一个发展中国家,在相当长的一段时间内还会面临着严重的交通安全问题。EXCEL是一种简便、易操作的计算机软件,可以利用EXCEL的数据分析功能,求出道路交通事故中某些推测数据与多因素之间的比例关系,对道路交通事故分析有所帮助。     

表达新城疫病毒F基因的重组鸡痘病毒实时荧光定量PCR检测方法的建立

采用荧光染料SYBR Green渗入法,通过对real-time PCR反应条件进行优化,建立了real-timePCR检测方法,用该方法定量分析新城疫病毒(NDV)F基因在重组鸡痘病毒(rFPV-F-VP0)第1、5、10、15、20代次中的表达整合情况。结果表明,建立的标准曲线呈现良好的重复性和特异性,与模板浓度呈现良好的线性关系。在线性浓度范围内,随着模板量的减少,其对应的Ct值相应增大,0.99<相关系数(r2)<1,0.8<扩增效率(E)<1.2,熔解曲线为单一特征峰型。本试验精确定量了外源基因在重组鸡痘病毒中的表达水平,为阐明重组鸡痘病毒的表达机理提供了理论基础。     

The Ge.F.I. DNA Proficiency Test: Year-one experience

The Ge.F.I. (the Italian Speaking Working Group of ISFG) has launched the first DNA Proficiency Test in 2012. The aim is to increase the external quality controls in forensic genetics in Italy and to drive to the standardizations of methods within the laboratories. Reference and mixed stains typing as well as statistical exercises were proposed and 26 laboratories submitted results.
