We analyzed 17 Y-STR loci (DYS19, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS385a/b, DYS438, DYS439, DYS456, DYS458 and DYS464a/b/c/d) in 252 Japanese males using three multiplex PCR typing systems. Two variants were found at DYS385a/b. A total of 244 different haplotypes were observed, of which 239 were found in single individuals. The haplotype diversity for the 17 loci was 0.996. 相似文献

In order to identify polymorphic positions and to determine their frequencies and the frequency of haplotypes in the human mitochondrial control region, two hypervariable regions (HV1 and HV2) of the mitochondrial DNA (mtDNA) of 374 unrelated individuals from Slovakia were amplified and sequenced. Sequence comparison led to the identification of 284 mitochondrial lineages as defined by 163 variable sites. Genetic diversity (GD) was estimated at 0.997 and the probability of two randomly selected individuals from population having identical mtDNA types (random match probability, RMP) for the both regions is 0.60%. 相似文献

In recent years it has been established that non-coding variants may be in linkage disequilibrium (LD) with coding variants up to several thousand base pairs away forming haplotype blocks. These non-coding markers may be haplotype specific and, therefore, informative regarding the surrounding coding sequence. In this study, we chose to study the VWA short tandem repeat (STR) as it is targeted in all major commercial kits utilized in routine forensic DNA profiling and is located in the von Willebrand Factor (vWF) gene; a gene associated with von Willebrand's Disease (vWD). We examined the VWA STR together with single nucleotide polymorphisms (SNPs) located throughout the vWF gene to identify haplotype structures and the extent of LD between markers in the region. Several areas exhibiting LD were identified by population data analysis in the 178 kilobase (178kb) vWF gene, which was supported by family studies. However, there appeared to be no evidence of LD blocks surrounding the VWA STR and evidence for recombination within 3 kb of VWA, hence, it is unlikely that VWA STR alleles could be used to predict haplotypes within the vWF gene that are associated with different forms of vWD. 相似文献

Haplotypes and allele frequencies of 17 Y-chromosomal short tandem repeat (Y-STR) markers were examined using the AmpFlSTR Yfiler PCR Amplification Kit (Applied Biosystems) in a population sample of 1166 Japanese male volunteers in 6 prefectures: Miyagi, Yamagata, Osaka, Tottori, Fukuoka, and Okinawa. A total of 1058 haplotypes were observed from 1166 males, and the most common haplotype detected in 12 males had a frequency of 1.03% and the discrimination capacity was 0.907. The RST analysis showed statistically significant differences between Okinawa and the other subpopulations. 相似文献

Mitochondrial DNA sequences of the entire control region were analyzed in 200 unrelated individuals from Macedonia. A total of 163 different haplotypes were found as determined by 177 polymorphic sites. The probability of a random match was calculated as 1:121 (0.83%). The basic phylogenetic structure of the Macedonian population as derived from its haplogroup distribution is in agreement with other West-Eurasian populations. Within an Italian collaborative exercise on the extended haplotype of the Y-chromosome, 1288 subjects were typed by the AmpFlSTR YFiler Amplification Kit (AB Applied Biosystems) and other 526 were typed by the PowerPlex Y® System (Promega). The sampling scheme included either a “regional” or a “local” recruitment, the first referring to individuals born in the region of the participating lab, the second referring to individuals coming from small villages. Total sample sizes were N = 954 and 860, respectively. A significant decrease of haplotype diversity was found in the local samples. The results may be of interest in forensic applications of the Y-chromosome. 相似文献

Two hundred and fifty-two different haplotypes were observed. The most common three haplotypes were shared by 0.8% of the sample, respectively.
