We report the use of immunohistochemical staining for analysis of forensic evidence from a double homicide. A 38-year-old woman and her 7-year-old daughter were murdered by multiple blows to the head and face with a tomahawk, resulting in multiple fragments of brain tissue scattered about the murder scene. The victims' husband and father was the main suspect, who maintained that he was out of town on business during the evening of the murders. However, a shirt taken from the suspect's car on the morning after the murders (secured by the police before the suspect visited the murder scene) was found to have two small stains. DNA analysis on the stains showed the presence of the deceased wife's DNA, and immunohistochemical stains on shirt fragments conclusively documented the presence of deep central nervous system tissue, providing the critical piece of evidence needed to arrest and prosecute the suspect. This report demonstrates that shirt or similar cloth fragments can be processed into paraffin blocks and subsequently immunostained to search for and classify types of tissue fragments that may be present on the fabric.  相似文献

Post-mortem interval (PMI) information sources may be subject to varying degrees of reliability that could impact the level of confidence associated with PMI estimations in forensic taphonomy research and in the practice of medico-legal death investigation. This study aimed to assess the reliability of PMI information sources in a retrospective comparative analysis of 1813 cases of decomposition from the Allegheny County Office of the Medical Examiner in Pittsburgh, US (n = 1714), and the Crime Scene Investigation department at Southwest Forensics in the UK (n = 99). PMI information sources were subjected to a two-stage evaluation using an adapted version of the 3x5 aspects of the UK police National Intelligence Model (NIM) to determine the confidence level associated with each source. Normal distribution plots were created to show the distribution frequency of the dependent variables (decomposition stage and source evaluation) by the independent variable of PMI. The manner, location, and season of death were recorded to ascertain if these variables influenced the reliability of the PMI. A confidence matrix was then created to assess the overall reliability and provenance of each PMI information source. Reliable PMI sources (including forensic specialists, missing persons reports, and digital evidence) were used across extensive PMI ranges (1 to 2920 days in the US, and 1 to 240 days in the UK) but conferred a low incidence of use with forensic specialists providing a PMI estimation in only 35% of all homicide cases. Medium confidence PMI sources (e.g., last known social contact) accounted for the majority of UK (54%, n = 54) and US (82%, n = 1413) cases and were associated with shorter PMIs and natural causes of death. Low confidence PMI sources represented the lowest frequencies of UK and US cases and exclusively comprised PMI information from scene evidence. In 96% of all cases, only one PMI source was reported, meaning PMI source corroboration was overall very low (4%). This research has important application for studies using police reports of PMI information to validate PMI estimation models, and in the practice of medico-legal death investigation where it is recommended that i) the identified reliable PMI sources are sought ii) untested or unreliable PMI sources are substantiated with corroborating PMI information, iii) all PMI sources are reported with an associated degree of confidence that encapsulates the uncertainty of the originating source.  相似文献

This study evaluated the use of different degree day (DD) models, developmental thresholds and developmental data sources for estimating postmortem interval (PMI) based on developmental rates of blow flies (Diptera: Calliphoridae). Blow fly evidence was collected from three mock crime scenes and sent for blind analysis of PMI. PMI estimates were calculated using averaging, single sine, double sine, single triangle, and double triangle models of DD calculation with minimum developmental thresholds of 0, 6 and 10 °C and Anderson's, Byrd and Allen's, Greenberg's, Nishida's and Kamal's developmental data sets. These estimates were compared with the actual PMI to determine which combination of DD model, developmental threshold and developmental data provided the most accurate estimate. For all three cases, at least one method gave a PMI estimation that was within 1 day of the actual date of death. There was no variability between PMI estimated using the five models of DD calculation. Overall, as lower developmental threshold increased, the PMI interval estimates increased. Depending on the developmental data set used, increasing the lower developmental threshold caused some overestimations of the date of oviposition, with estimates given that were prior to the actual death. Future PMI estimates should include error ranges, so that overestimation of PMI is avoided.  相似文献

The authors have published elsewhere a quantitative method for assessing weight of evidence in the case where a finger mark from a crime scene is compared with a control print taken from a single finger of a suspect. The approach is based on the notion of calculating a likelihood ratio (LR) that addresses a pair of propositions relating to the single finger that was the origin of the crime mark. In practice, things are rather different because the crime mark will not just be compared with a single finger from a suspect but with a set of prints from all of his/her fingers; likewise, when the mark is compared with a database, this will consist of ten print records from random individuals. It is clear that "finger propositions" are not realistic in this situation and we show how our approach may be generalised to address a pair of propositions that relate to the person that made the crime mark. It often is the case that information is present at the crime scene that enables some inference to be drawn relating to which of the offender's ten fingers left a particular mark of interest. This kind of inference may profitably be drawn into the formal analysis. We illustrate our approach with an example.  相似文献

Images and videos are common types of evidence in crime scene investigations and laboratory analysis. Images may be taken by the suspect and/or by crime scene investigators and may serve as crucial elements in forensic laboratory analysis. Forensic photographic image comparison is the process of comparing one or more objects or persons in photographs/images when at least one image is known to be related to a crime. The forensic examiner usually compares the images in order to determine whether or not an association between the exhibits in the images can be made. This paper proposes an extension to the currently prevalent photographic image examination method. The extension introduces comparison of landscape and vegetation over time. It is revealed that similarities between images may still be found between the period of time the suspect photograph was taken and the period the crime scene investigator took the photograph from the same area. In this case report, two suspects to be involved in growing a marijuana field were arrested by the police. The forensic experts were asked to examine images taken by the crime scene investigators and to compare them to the images found in the suspects’ phones. They then tried to determine whether the suspects could be linked to the specific locations. While applying techniques derived from morphological comparison methodologies, the plants at the scene provided significant additional information. A tree trunk, branches, and twigs on a hedge in the photographs revealed specific individual characteristics that led the examiner to reach a conclusive decision.  相似文献

A set of historic murders, known as the “Jack the Ripper murders,” started in London in August 1888. The killer's identity has remained a mystery to date. Here, we describe the investigation of, to our knowledge, the only remaining physical evidence linked to these murders, recovered from one of the victims at the scene of the crime. We applied novel, minimally destructive techniques for sample recovery from forensically relevant stains on the evidence and separated single cells linked to the suspect, followed by phenotypic analysis. The mtDNA profiles of both the victim and the suspect matched the corresponding reference samples, fortifying the link of the evidence to the crime scene. Genomic DNA from single cells recovered from the evidence was amplified, and the phenotypic information acquired matched the only witness statement regarded as reliable. To our knowledge, this is the most advanced study to date regarding this case.  相似文献

In a case of alleged sexual assault, the pollen content of samples of grass clippings and soil from the suspect's clothing and shoes was compared to that of a sample of grass clippings from the alleged crime scene (a grassy area) to determine whether or not the suspect had been at the scene. The clothing and shoe samples showed a very strong correlation with each other and with the sample from the alleged crime scene in the combination of the different types of pollen present, very strongly supporting the contention that the suspect had been at the scene.  相似文献

Criminal cases are studied from several disciplines to link a suspect with a criminal act. In this case, a man was reported missing in a coastal area in Buenos Aires Province, Argentina. The victim's relatives pointed to a possible suspect, and the local police carried out the investigation. We contributed to this research by applying palynological and mycological techniques. Palynomorphs and fungal spores offer valuable trace evidence, as they can be easily transferred between objects and crime scenes due to their minute size and persist on them for a long time. The victim was found 25 days later, lying on sandy soil, which partially covered the body, 35 km from where the suspect was arrested. Comparative samples were collected from the crime scene and the suspect's home and belongings (clothes, footwear, and seized vehicle). The palynological associations obtained from the crime scene and the defendant's belongings were dominated by diatoms and acritarchs (Acantomorphitae), all elements of marine origin, and a high CFU number of Bipolaris cynodontis, which allowed the defendant's clothing to be related to the place of corpse discovery. Soil from the defendant's home had an entirely continental composition, and the fungal biota was characteristic of prairie areas which were different from those of the crime scene.  相似文献

The association between insect development and temperature is well established. Thermal summation using accumulated degree-day measures is commonly used. However, the time at which evidence is collected is important in these estimates. The aim of this study was to provide a simulated model of the effect of temperatures on six dipteran species commonly associated with cadavers, from the death scene to the refrigerator, and finally at the time of autopsy. Temperatures measurements were sampled over a 16-month period from the external environment (external to the mortuary), within the mortuary refrigerator, and within the mortuary autopsy suite. Monte Carlo simulation using accumulated degree-days (ADD) was used to estimate the variations based on the mean and standard deviation of the temperature measurements. It was found that there was a negative correlation between the base temperature of the fly species (lowest temperature at which the flies will survive) and developmental likelihood. Species with high base temperatures (Chrysomya albiceps, Chrysomya chloropyga, and Musca domestica) were less likely to continue development in refrigerators than species with lower base temperatures (Lucilia sericata and Piophila casei). The findings of this study highlight the importance of recording temperature measurements and the period of refrigeration on PMI estimation especially when continued development occurs in spite of a period of cooling of the insect evidence.  相似文献

Species identification of necrophagous insects found on a dead body is an essential key in applying medicolegal entomology to the estimation of postmortem interval (PMI). Due to limited morphological identification of insect evidence, several studies have identified species using molecular information such as DNA markers. While considerable cytochrome c oxidase subunit I (COI) gene sequence data of necrophagous fly species have been collected and annotated, those of necrophagous beetle species have not. Since necrophagous beetles such as Dermestes species have a larval period longer than that of flies, beetles are useful in even the late decomposition phase in estimating minimum PMI. To obtain the full-length COI gene sequences of six Dermestes species collected from South Korea, we designed primers for polymerase chain reaction amplification and sequencing. The obtained full COI nucleotide sequences were used for performing phylogenic analysis and comparison with previously reported sequences. The results demonstrated that the COI gene sequences could be used to identify forensically important Dermestes species in South Korea.  相似文献

In the investigation of arson, evidence connecting a suspect to the fire scene may be obtained by comparing the composition of ignitable liquid residues found at the crime scene to ignitable liquids found in possession of the suspect. Interpreting the result of such a comparison is hampered by processes at the crime scene that result in evaporation, matrix interference, and microbial degradation of the ignitable liquid. Most commonly, gasoline is used as a fire accelerant in arson. In the current scientific literature on gasoline comparison, classification studies are reported for unevaporated and evaporated gasoline residues. In these studies the goal is to discriminate between samples of several sources of gasoline, based on a chemical analysis. While in classification studies the focus is on discrimination of gasolines, for forensic purposes a likelihood ratio approach is more relevant. In this work, a first step is made towards the ultimate goal of obtaining numerical values for the strength of evidence for the inference of identity of source in gasoline comparisons. Three likelihood ratio methods are presented for the comparison of evaporated gasoline residues (up to 75% weight loss under laboratory conditions). Two methods based on distance functions and one multivariate method were developed. The performance of the three methods is characterized by rates of misleading evidence, an analysis of the calibration and an information theoretical analysis. The three methods show strong improvement of discrimination as compared with a completely uninformative method. The two distance functions perform better than the multivariate method, in terms of discrimination and rates of misleading evidence.  相似文献

DNA forensics and the poaching of wildlife in Italy: a case study   总被引:2,自引:0,他引:2
DNA molecular techniques were used in a forensic investigation involving the poaching of wildlife in a national park of Italy. A poacher, after having snared a wild boar (Sus scrofa) sow, knifed it to death. The animal was retrieved by conservation officers at the scene before the poacher could remove the carcass. Subsequently, the suspect denied the charges. During a search of his home, a bloodstained knife was confiscated. A method to identify the species from the DNA extracted from the stains revealed the blood to be that of the non-domestic form of Sus scrofa. Further DNA typing for individual identity using species-specific single tandem repeats or microsatellites (STRs) showed that the DNA on the knife matched that of the poached boar. Based upon the forensic evidence obtained, the suspect was convicted of poaching and of cruelty to animals.  相似文献

When insect evidence is obtained during autopsy, forensic entomologists make decisions regarding the effects of low-temperature (-1 degrees C to 4 degrees C) storage of the body and associated insects when estimating the post-mortem interval (PMI). To determine the effects of storage in a morgue cooler on the temperature of maggot masses, temperatures inside and outside of body bags containing a human cadaver and porcine cadavers (seven replicates) were measured during storage. Temperatures remained significantly higher (p<0.05) inside of the body bags relative to the cooler, and remained at levels sufficient for maggot feeding and development. If the assumption that no insect development takes place during preautopsy refrigeration is made, potential error rates in PMI estimation of 8.6-12.8% occur. The potential for blow fly larvae to undergo significant development while being stored in the morgue is a possibility that forensic entomologists should consider during an investigation involving samples collected from autopsy. Case and experimental evidence also demonstrate that substantial tissue loss can occur from maggot feeding during morgue storage.  相似文献

A young woman was found unconscious in her flat strangulated with a children's scarf and was at first thought to be dead. The victim survived but is disabled for life. Neither bloodstaines nor sperm were found at the scene. Swebs of fleece were prepared from two fingers of a one-way glove which were found at the scene and from each end of the scarf in order to find some epithelial cells from the criminal. DNA was extracted and amplified by the polymerase-chain-reaction (PCR). The DNA-profile of the victim as well as that of the suspected apart living husband were found in these mixed stains. The suspect was severely incriminated by these findings and other evidence. Consequently the court convicted him to eleven years of inprisonment because of attempted manslaughter.

The use of temperature-based short-term postmortem interval (PMI) estimation methods can be useful to homicide investigators at the scene of a questionable death; however, a number of current PMI estimation techniques have high error rates such that they are of limited utility to law enforcement investigators and forensic professionals. These methods fail to control confounding errors present in individual data. An averages-based method of short term postmortem interval estimation was compared to eight other methods, and found to predict postmortem interval considerably more accurately, and for much longer periods of time.  相似文献

Sexual assault casework requires the collaboration of multiple agency staff to formalise an investigative pipeline running from crime scene to court. While the same could be said of many other forensic investigations, few require the additional support of health care staff and the combined forensic involvement of body-fluid examiners, DNA experts and analytical chemists. The sheer amount of collaborative effort between agencies is laid out through a detailed examination of the investigative workflow from crime scene to courtroom with each step in the pipelines detailed and discussed. Beginning with a review of sexual assault legislation in the United Kingdom this article details how sexual assault investigations are initiated by police and supported by sexual assault referral centre (SARC) staff who are often the first responders providing primary healthcare and patient support to victims while simultaneously collecting and assessing forensic evidence. Detailing the myriad of evidential material that can be documented and collected at the SARC, the review identifies and categorises key forensic tests to first detect and identify body-fluids recovered from evidence through to the secondary analysis of DNA to help identify the suspect. This review also focusses on the collection and analysis of biological material used to support the allegation that the sexual activity was non-consensual and provides a breakdown of common marks and trauma as well as a review of common analytical methods used to infer Drug Facilitated Sexual Assault (DFSA). The culmination of the investigative pipeline is discussed by reviewing the Rape and Serious Sexual Assault (RASSO) workflow used by the Crown Prosecution Service before providing our thoughts on the future of forensic analysis and possible changes to the described workflows.  相似文献
