Abstract:  With <100 pg of template DNA, routine short tandem repeat (STR) analysis often fails, resulting in no or partial profiles and increased stochastic effects. To overcome this, some have investigated preamplification methods that include the addition of proofreading enzymes to the PCR cocktail. This project sought to determine whether adding proofreading polymerases directly in the STR amplification mixture would improve the reaction when little template DNA is available. Platinum Taq High Fidelity and GeneAmp High Fidelity were tested in Profiler Plus? STR reactions alone and in combination with AmpliTaq® Gold. All reactions included the additional step of a post‐PCR purification step. With both pristine low template DNA and casework samples, the addition of these polymerases resulted in comparable or no improvement in the STR amplification signal. Further, stochastic effects and artifacts were observed equally across all enzyme conditions. Based on these studies, the addition of these proofreading enzymes to a multiplex STR amplification is not recommended for low template DNA work.  相似文献

In this study we aim to compare the public perceptions towards informal patient payments in six Central and Eastern European countries (Bulgaria, Hungary, Lithuania, Poland, Romania and Ukraine). Overall, around 35–60% of the general public in each country has ever made informal payments, though informal cash payments are perceived negatively, mostly as corruption. In-kind gifts are often seen as a token of gratitude. However, significant differences among countries are observed. Despite the public support for the eradication of informal payments, there are population groups who favor their existence and this should be taken into account in policy-making.  相似文献

Outside the health care sector, consumer preferences have been effectively studied using rating and ranking conjoint techniques. In the health care sector this technique has received less attention than its choice-based variant. Applications of rating and ranking method to health care issues are few. This paper presents an application of rating conjoint analysis to study the importance of quality, access and price to the health care consumers in Bulgaria. The paper first describes the rating conjoint method and its distinctive features compared to the choice-based and the ranking approach. The method is illustrated by the rating conjoint design applied in the study. Next, the impact of different quality-, access- and price-levels on the rating of physician profiles is analysed and the differences between the socio-demographic groups are examined. The results suggest that similar to other countries, the quality of care is a highly valued characteristic in Bulgaria, whereas access is perceived as less important. The considerable importance of patient payments further implies that Bulgarians are responsive to prices in the health care sector, especially the elderly, the village dwellers and the lowest income groups. The relevance of the results with regards to health policy and planning, as well as with regards to the methodology of rating conjoint analysis is discussed at the end of the paper.  相似文献

Contemporary democracies show considerable differences in the issue composition of their protest politics, which tends to remain relatively stable over time. In countries like Germany or the Czech Republic, the vast majority of protests have been mobilised around sociocultural issues, such as human rights, peace, nuclear power or the environment, and only a tiny portion of protest has focused on economic issues. At the opposite extreme, protest in France or Poland usually has a strongly economic character and voices demands relating to material redistribution and social policy. What lies behind the cross-country differences in national protest agendas? In this article, the national protest agenda depends on what issues mainstream political parties are contesting: the content and strength of the master-issue dimension. In reference to the literature on the multidimensional political space and niche political parties, one should expect that there is a substitutive effect; where the stronger a specific master-issue dimension is in party politics, the less salient that issue dimension is in protest politics. This substitutive effect results from the tendency of electoral politics to reduce political conflict to a single-dimension equilibrium, which decreases the importance of other issues and relegates the contest over secondary, niche issues to the realm of policy-seeking strategies, with protest being a common type of this political strategy. In party systems where single-dimension equilibrium does not exist and the master-issue dimension is weaker, the same dynamics result in a more convergent relationship between party and protest politics and a greater similarity between the protest- and party-system agendas. To investigate this theory, the national protest agendas in four countries are examined. The Czech Republic, Hungary, Poland and Slovakia show four combinations of two crucial factors that are not available in the old Western democracies: the content and the strength of the master-issue dimension. The study draws on an original dataset of protest events organised in the four countries between 1993 and 2010, and on qualitative and quantitative data on issue dimensions of party politics obtained from studies on party politics and expert surveys. The results show that in the Czech Republic, where the master-issue dimension has remained strongly economic, protest has been predominantly sociocultural. In Poland between 1993 and 2001 and Hungary between 1993 and 2006, the master-issue dimensions are strongly sociocultural, while protest is predominantly economic. There is no single-dimension equilibrium in party politics in Slovakia or in post-2001 Poland and mainstream parties compete on both economic and sociocultural issues. Consequently, the substitutive dynamics between party and protest politics is weaker and the issue agendas in party and protest arenas are here more alike.  相似文献
