猪繁殖与呼吸综合征病毒在猪动脉血管内皮细胞上的培养特性

探索应用原代猪血管内皮细胞培养猪繁殖与呼吸综合征病毒(PRRSV)的方法,为进一步研究PRRSV对血管内皮细胞的损伤及其作用机制建立模型。用Ⅱ型胶原酶灌注60日龄健康仔猪颈总动脉分离获得动脉血管内皮细胞,以PRRSVHuN4株接种原代猪血管内皮细胞,进行细胞形态和荧光定量RT-PCR检测。结果显示,PRRSV接种原代猪血管内皮细胞后,呈现典型的细胞病变,病毒增殖规律与Marc-145细胞基本相同。结果表明,在猪血管内皮细胞上成功复制了PRRSV,为下一步研究PRRSV在血管内皮细胞中的复制及其导致的病理损伤提供了试验依据。     

猪卵丘细胞和胎儿成纤维细胞的分离培养及传代

对猪卵丘细胞和胎儿成纤维细胞的分离、培养和传代进行了系统研究。结果显示,猪的卵丘细胞生长形成单层需要5~6 d;运用组织块法和酶消化法获得胎儿成纤维细胞单层的生长时间分别为10~11 d和8~9 d。猪的卵丘细胞和胎儿成纤维细胞均可以建立并获得稳定的细胞系,这些方法的应用可为猪体细胞核移植研究提供丰富的供体细胞。     

猪胚胎多能性干细胞的分离培养

收集妊娠 2 6 .0～ 2 9.0d的猪胚胎 ,分离培养原始生殖细胞 (PGCs) ,在STO细胞饲养层上生长 ,形成了多能性干细胞 (EG)。发现其具有干细胞的显著特性 ,并能在体外定向分化为神经细胞、上皮细胞和胚泡细胞。试验使用A、B、C 3种不同的培养基培养PGCs ,结果在形成EG细胞集落的数量和EG细胞分化上有着明显的差距 ,说明血清的质量和浓度以及细胞因子对干细胞的生成和增殖有着重要的影响     

低血清培养PK-15细胞系对TGEV增殖规律的研究

为降低疫苗生产综合成本,本研究探讨了以低血清培养基(M08011)替代常规细胞培养基(DMEM)的可行性。本研究依次采用含100、80、50、30、20、10、0mL/L血清的低血清培养基(M08011)驯化PK-15细胞,并考察猪传染性胃肠炎病毒(TGEV)在低血清培养的PK-15细胞系中的增殖规律。结果显示,驯化后的PK-15细胞在含100、80、50、30、20 mL/L的低血清培养基中生长状态良好,与常规培养基所培养的PK-15细胞无差异。低血清培养体系培养PK-15细胞时,血清最低添加量为20 mL/L。用该体系培养的PK-15细胞接种TGEV后,TGEV的TCID50为10^-4.97/100μL,与常规条件培养的TGEV的TCID50为104.88/100μL相比,差别不明显。TGEV接种PK-15细胞,低血清培养基中血清浓度降低至20 mL/L,毒价未受到影响。结果表明,本研究建立的PK-15细胞及TGEV低血清培养体系可初步应用于TGEV的增殖培养,为相关疫苗研发奠定了基础。     

猪用生长激素脂质体对大白鼠的促生长活性试验

对大白鼠分别腹腔注射猪用生长激素 (PST)和PST脂质体 ,第 7、14、2 8d称重 ,试验组与对照组之间差异显著 (P <0 .0 5 ) ,表明PST和PST脂质体对大白鼠具有促生长活性 ;同时显示 ,PST脂质体具有显著的缓释作用 ,缓释期可达 7d以上。     

八珍汤对乳牛产后免疫状态的影响

为了探索提高乳牛产后期免疫状态的新方法,对试验乳牛分4组(产前第30 d至产前第1 d灌喂组、产前第15 d至产后第15 d灌喂组、产后第1 d至第30 d灌喂组、不灌喂中药对照组)灌喂中药八珍汤,检测产后期淋巴细胞及其亚群数量和淋巴细胞增殖活化功能。结果发现,产前组CD3细胞数量在产后第1 d升高;产前产后组CD3、CD4和CD8细胞数量在产后第1 d和第15 d升高;产后组CD3细胞数量在产后第15 d和第30 d升高,CD4细胞数量也在产后第30 d升高。淋巴细胞对ConA的反应能力,产前产后组在产后第1~30 d明显提高,产后组在产后第15 d和第30 d明显提高。各组乳牛产后期CD21细胞数量的变化相近。结果表明,从产前第15 d开始到产后第15 d每日喂八珍汤,能明显提高乳牛产后期T细胞及其亚群数量和增加淋巴细胞增殖活化功能,而对B细胞数量增加的作用不明显。     

牦牛胎儿成纤维细胞的分离培养

采用30d胚龄左右的牦牛胚胎,用2种不同的消化液消化后,将所得的牦牛胎儿成纤维细胞(yak embryonic fibroblasts,YEF)培养于不同血清含量的RPMI1640培养液中,并接种于2种不同材质的培养瓶表面,对YEF的分离方法、培养所需胎牛血清(FBS)添加量和不同基质表面对YEF生长的影响进行了研究。结果表明,含150 mL/L FBS的RPMI1640培养基最适合YEF生长,可传代27次以上;经用RPMI1640和D-Hank’s液配制的2．5 g／L胰蛋白酶消化,所得的YEF的细胞活率差异不显著(P>0．05),其原代细胞在含100 mL／L胎牛血清的RPMI1640培养液中的贴壁率差异极显著(P<0．01);不同生长基质对YEF的形态没有显著影响。     

人工感染PRRSV仔猪肺和子宫细胞凋亡的电镜观察

应用透射电子显微镜对仔猪感染猪生殖与呼吸综合征病毒(PRRSV)后不同时期肺和子宫凋亡细胞的超微形态结构进行了观察。结果表明,PRRSV可诱导宿主肺和子宫发生典型的细胞凋亡,表现为细胞的超微形态结构随着病毒感染进程出现不同的特征性变化,早期(感染后第8 h至第3 d)凋亡细胞多表现为细胞体积缩小,细胞质密度增强,染色质浓缩,核仁解体,内质网扩张;中期(感染后第5 d至第9 d)凋亡细胞体积显著缩小,细胞膜突起,线粒体增多,有凋亡小体形成;晚期(感染后第10 d以后)凋亡细胞形态多表现为凋亡小体降解和消失,少数凋亡细胞在凋亡晚期表现为坏死细胞。肺细胞凋亡主要发生于肺巨噬细胞和肺泡上皮细胞,子宫细胞凋亡主要发生于子宫内膜细胞和内膜腺细胞。     

乳牛产后期生理性免疫状态的研究

为了研究乳牛在产后期的生理性免疫状态,检测了产后期乳牛淋巴细胞及其亚群数量和淋巴细胞增殖活化功能及其变化。结果发现,乳牛淋巴细胞及其亚群数量和淋巴细胞增殖活化功能在分娩后第1 d最低,然后表现出缓慢上升的总体趋势。CD3细胞数在产后第15 d低于产后第1 d,到产后第30 d时达到最高值;CD4细胞数一直多于CD8细胞数;CD21细胞数在产后第15 d时达到最高值,以后缓慢下降;淋巴细胞的增殖活化能力在产后第30 d最高,未观察到淋巴细胞有明显的凋亡和增生现象。结果表明,乳牛免疫状态在产后第1 d最低,以后逐渐升高,第15 d时细胞免疫最低而体液免疫最高,第30 d时细胞免疫最强。     

对《猪食盐中毒的治疗》一文治疗方法的不同看法

笔者读《中国兽医科技》1988年第2期19页“猪食盐中毒的治疗”一文后,觉得该文介绍的治疗方法有些不妥,现谈谈个人浅见,以求同道指正。 猪对食盐极为敏感,过多的食盐进入胃肠被吸收后容易发生食盐中毒,一但出现中毒,死亡率很高。其毒理是:食盐在体内分解使钠离子浓度过高,易形成钠、钾优势,从而产生以神经反射亢进的临床症状,如肌肉痉挛、抽搐等。进而由于大量的氯化钠进入血液,使细胞外液渗透压迅速增高,细胞内液流向细胞     

The Development of EU-Russian Relations

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. But the two sides would refrain from extreme measures and prevent their relations from sliding into real cold war.     

Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.
