雏鹅新型病毒性肠炎病毒对鸭胚成纤维细胞的适应性及增殖特性

为了获得适宜雏鹅新型病毒性肠炎病毒(NGVEV)体外培养的细胞系,开展了NGVEV强毒株在鸭胚成纤维细胞(DEF)适应性和增殖特性的研究。结果表明,NGVEV强毒株经尿囊腔接种10日龄鸭胚传4代,取尿囊液接种DEF,第1代无明显细胞病变;第2代培养至72~96 hDEF出现颗粒状缺失、脱落,但无典型蚀斑出现;第3代培养至96~120 h,DEF形成大小不等、圆形或椭圆形的典型蚀斑;第5代以后的NGVEV可稳定适应于DEF,典型细胞病变出现于48~72h。取适应DEF的NGVEV感染DEF细胞后制作超薄切片经电镜观察,可见典型的腺病毒粒子,大小为70~90 nm。NGVEV在DEF上的TCID50值随着传代次数增加而增高,由第1代的101.23增加到第8代的108.02,最高毒价出现于96~144 h,96~120 h是收获病毒的最佳时间。     

禽腺病毒血清4型河南株的分离鉴定及对鸡胚致病性的研究

为探明河南省某鸡场疑似禽腺病毒血清4型发病鸡群的感染情况和病毒特征,本试验对发病鸡群进行了病毒分离鉴定,并研究所分离到的病毒对鸡胚的致病性。采集发病鸡群鸡肝,处理后经尿囊腔接种SPF鸡胚,尿囊液禽腺病毒血清4型PCR鉴定和hexon基因序列分析,尿囊液经过浓缩定量和梯度稀释,接种SPF鸡胚,观察鸡胚病变,计算ELD_(50),同时荧光定量PCR测定病毒在鸡胚肝和尿囊液中的载量。结果显示,在鸡胚尿囊液中扩增出大小934 bp的基因片段,序列分析表明,该序列与禽腺病毒血清4型同源性为98.2%~99.8%。感染病毒的鸡胚的胚体矮小、发育不良,肝肿大、质脆,呈黄色或棕黄色,胸肌、腿肌明显出血,病毒载量和鸡胚病变与接种剂量有关。该毒株的ELD_(50)为1.36×10~7copies/L,相同时间内病毒在肝中的载量高于尿囊液。本试验为进一步开展禽腺病毒血清4型的研究和防控提供了必要的参考依据。     

乌鸡新城疫病毒中发型毒株的分离与鉴定

从发病的 43日龄乌鸡群中分离到 1株病毒 ,经鸡胚传代培养后 ,测定其F3 代尿囊液血凝 (HA)效价为 2 8,HI试验中能被鸡ND阳性血清中和 ,鸡胚平均死亡时间 (MDT)为 60 .8h ,1日龄雏鸡脑内接种指数 (ICPI)为 0 .8,血凝解脱时间小于 10min ,血凝素热稳定性差。表明该分离株为新城疫病毒中发型毒株。     

抗小鹅瘟免疫血清和免疫卵黄浆的制备及临床应用效果

(一)材料与方法 1. 种毒和疫苗:种毒系江苏农学院牧医系微生物组提供的SYG_(61)代毒;疫苗为本站从用SYG_(61)代毒接种并产生典型病变的鹅胚收获的尿囊液。 2. 供试鹅及种蛋:从市场购入体重为2～3.5kg健康成年菜鹅191只;3日龄商品雏鹅由市郊炕坊提供;制卵黄浆用的蛋为经两次主动免疫的母鹅在免疫后1个月所产的新鲜蛋。 3. 血清的制备:①免疫程序:用上述尿囊液10~(-2)×1ml肌肉注射,进行基础免疫;隔     

鸡肾型传染性支气管炎病毒的分离及其N基因片段的克隆和测序

通过形态学观察、动物回归试验、鸡胚接种试验、病毒干扰试验以及血凝试验分离鉴定了1株肾型鸡传染性支气管炎病毒(IBV)。在透射电子显微镜下,病毒粒子多呈球形,直径为80~120 nm,有囊膜,表面有冠状突起。鸡胚连续盲传至第 1~2 代,开始出现死亡或出现侏儒胚;分离株可显著干扰NDV在鸡胚中的增殖;病毒尿囊液无凝血活性,但经5 g/L胰蛋白酶处理后,能够凝集10 mL/L的鸡红细胞。利用RT PCR技术对分离株的N基因进行了扩增,经克隆、序列测定和分析比较,证实分离株为肾型IBV,命名为AH3 04株。     

鸭肿头出血症病毒在鸭胚原代成纤维细胞中增殖特性的研究

将鸭肿头出血症病毒(DSHDV)强毒株经尿囊腔接种10日龄鸭胚,传3代,取第3代尿囊液接种鸭胚成纤维细胞(DEF),传代培养至有细胞病变出现,并对接毒细胞进行TCID50动态检测和透射电镜观察。结果显示,第1代接毒细胞培养至120 h出现变圆、脱落,并形成少量蚀斑,第2代培养至96~120 h,DEF形成大量圆形或椭圆形的典型蚀斑,第3代培养至72~96 h,DEF出现典型蚀斑;此后可稳定适应于DEF,典型细胞病变出现于72~96 h。DSHDV在DEF上的TCID50值随传代次数的增加而增高,由第1代的102.75增加到第10代的106.82,最高毒价出现于96 h,该点是收获病毒的最佳时间。经透射电镜观察,可见DSHDV粒子呈球形或椭圆形,大小为60~75 nm,无囊膜,具有双层衣壳。结果证实,DSHDV强毒株已经适应了DEF,传代后可获得较高的病毒效价。     

鸡胚成纤维细胞培养鸭瘟病毒的试验

将鸭瘟病毒强毒(DPV34)经12日龄鸭胚连续传2代,收获的尿囊液DPV34F2作为细胞培养的病毒接种于鸡胚成纤维细胞,连续传代5次.结果,从第2代开始,接种DPV的鸡胚成纤维细胞出现细胞病变,随着代次的增加,细胞病变愈加明显.经电镜观察,第5代细胞培养物中有典型的DPV病毒粒子;将第5代培养物接种鸭胚,出现典型鸭瘟病变;用PCR方法检测培养物,也出现DPV的特征DNA带.     

腺胃型鸡传染性支气管炎病毒Hu98株的分离鉴定

1998年首次从哈尔滨市某鸡场鸡腺胃肿大为特征的病例中分离到IBV-Hu98毒株.将该病毒株接种SPF鸡胚并传至5代(F5),结果接种胚出现规律性死亡和典型胚胎病变特征,EID50为10-6.40/0.2 mL.IBV-Hu 98 F5感染SPF鸡胚尿囊液经负染电镜观察,可见直径为80～120 nm、有囊膜和纤突的冠状病毒粒子.用IBV-Hu98 F5毒株人工感染1日龄SPF鸡能引起与自然病例相似的病理组织学变化,死亡率为80%(4/5),用IBV-Hu 98J1人工感染1日龄SPF鸡死亡率为100%(5/5),并可从死亡鸡中分离到冠状病毒.     

免疫酶斑点法快速检测小鹅瘟病毒

至今,有关小鹅瘟病毒(GPV)的快速诊断尚未找到一种较为理想的方法,Malkinson(1974)和Peleg(1974)发现牛精子能被GPV所凝集,但为非特异反应,难以说明问题;Dosz-kow,K.I.等(1982)用酶组化法技术测定细胞培养和小鹅组织中的抗原;Bondarerke(1982)用免疫扩散技术测定鹅胚绒毛尿囊液中的抗原,然而,前者所用的抗体是常规血清,非特异性反应较强,后者需高效价血清及多倍浓缩的尿囊液,且灵敏度不高,这些方法在实际应用上均有一定的困难。1981年徐为燕等和1989年孙怀昌等用反向间接血凝技术检测鹅胚尿囊液及组织中的GPV,效果虽     

鸡病毒性关节炎病毒的分离与鉴定

将疑似病毒性关节炎病鸡关节炎炎性渗出物处理之后，在ＳＰＦ鸡胚上传代，第６代时鸡胚开始死亡，第９代以后呈现规律死亡，胚体出血，绒毛膜水肿。以此尿囊液接种Ｖｅｒｏ细胞出现明显的合胞体病变，继而产生严重的细胞脱落。电镜观察，病毒粒子呈六角形，无囊膜，直径约７０ｎｍ。琼扩试验中，该病毒与关节炎病毒特异抗血清呈明显阳性反应。回归试验中，该病毒能使１日龄ＳＰＦ鸡雏在７２ｈ内发生严重的关节炎病变，最终全部死亡。理化特性鉴定证明其核酸为ＲＮＡ，能抵抗乙醚和氯仿。结果表明该分离物为鸡病毒性关节炎病毒。

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.
