本研究利用真核表达系统表达非洲猪瘟病毒的CD2v胞外区蛋白,研究其免疫特性,制备多克隆抗体并研究其性质,为研发特异性单抗奠定数据基础。以非洲猪瘟病毒Wuhan2019-1株(GenBank:MN393476.1)为模板,合成CD2v胞外区基因序列,并将其克隆至真核表达载体pCAGGS载体中,获得pCAGGS-CD2v胞外区重组质粒。经酶切和测序鉴定后,将重组质粒转染293F细胞,获得重组胞外CD2v蛋白(简称His-CD2v)。用Ni-NTA柱纯化His-CD2v,并以此为抗原免疫BALB/c小鼠制备多克隆抗体,利用ELISA和间接免疫荧光对多克隆抗体进行鉴定。结果显示,(1)His-CD2v融合蛋白在293F中以可溶形式表达,SDS结果显示,相对分子质量约为35~80 ku的弥散带;(2)融合表达的His-CD2v能与阳性血清反应,与对照不反应;(3)HisCD2v制备的多抗效价最高达1∶218700,且能与阳性血清竞争性结合CD2v抗原。上述结果表明,His-CD2v融合蛋白的表达及其多抗的成功制备,为进一步筛选CD2v特异性单抗及研制竞争ELISA试剂盒,为非洲猪瘟感染与免疫鉴别方法的建立奠定了坚实的基础。 相似文献

This article analyses the formation, presentation and reception of two seminal exhibitions: Australian Painting: Colonial, Impressionist, Contemporary (1962–1963) and Canadian Painting 1939–1963 (1964). The presentation of these exhibitions at London’s Tate Gallery reflected the institution’s support for “old dominion” Commonwealth members. The exhibitions also highlight the differing visions of the Canadian and Australian governments concerning the relationship between art, diplomacy and politics during the Cold War. In Canada, Vincent Massey (Governor General 1952–1959) played a key role in ensuring that all forms of Canadian art were promoted internationally. Massey wanted to connect with the European and American avant-garde and to be part of a multiracial Commonwealth. This contrasted with the rather “old-fashioned” views of the Australian prime minister, Robert Menzies, and the Commonwealth Art Advisory Board. They supported a Commonwealth dominated by the “white dominions” and the initial exhibition plan for Australian Painting recalled previous British Empire art shows. The British response to the Canadian and Australian exhibitions is also discussed. British critics preferred the nationally identifiable “exotic” art found in Australian art to the transnational forms of international abstraction in Canadian art. This article focuses on the way the Anglophone Caribbean succumbed to the overhaul of the European Union sugar trade and how these countries have attempted to restructure their economies in its wake. We show how the protagonists of reform gave a sense of inevitability to the demise of the Commonwealth trade system and conveyed (unrealistic) strategies for how this should be managed for the benefit of the Caribbean. In this way we detail the hegemony of neoliberalism in contemporary trade politics and the need for alternative strategies for rural development in the Caribbean region. 相似文献

ABSTRACTThis paper examines the Commonwealth’s inclusive role in engaging with the distinctive challenges of education provision in small states, an agenda that is often neglected elsewhere. We examine the origins of Commonwealth work on education in small states, the nature of its comparative advantage, its role in facilitating small states’ engagement with international education dialogue and regional co-operation, and the demand and potential for ongoing Commonwealth support for education. Particular attention is given to experience within the Caribbean region and to the potential for the Commonwealth and the wider international community to learn from small states in the light of their distinctive educational challenges, achievements and priorities – and, most notably, their experience at the ‘sharp end’ of environmental uncertainty and climate change. 相似文献

Conscious policy and program support for the resettlement of refugees in regional and rural areas is a relatively recent trend in Australia. Resettlement is a complex process that hinges on the establishment of viable communities. This review suggests that its outcomes, especially for refugee communities, are potentially mixed. However, an appropriately resourced, well managed and well planned refugee resettlement program can potentially provide beneficial outcomes for refugees and host communities. This article draws on existing empirical data and contributions made at a VicHealth Roundtable in late 2007 to thematically explore the challenges facing rural and regional resettlement programs. Given that settlement is a dynamic process and the approaches to rural and regional settlement are evolving, this article provides 12 propositions for a more effective and integrated approach to policy and practice. 相似文献

This paper examines race-based peremptory challenges. Such challenges occur during the voir dire jury selection process. The process allows both the defense and the prosecution to strike jurors who they believe will not
decide cases fairly. However, in the case of Batson v. Kentucky 476 U.S. 79 (1986), the Supreme Court ruled that race could not be used as a factor in eliminating prospective jurors. This
paper examines federal litigation for five years in which it was alleged that race was used as a factor in removing a juror.
An examination of the cases revealed that most of the cases involved sole male litigants who allege that there were multiple
race-based peremptory challenges used in their cases. Moreover, most of the cases that led to the allegations involved violent
offenses. Other case characteristics are noted, but of most significance was the finding that most appellants lost their cases.
As such, the courts felt that most of the challenges were, in fact, race neutral. The implications of this research are discussed.

AbstractOf all the island states of the Southwest Pacific, Fiji’s foreign relations have been the most fraught since the advent of independence in the region, due largely to a succession of coups d'état. These have invariably precipitated adverse responses from major partners and aid donors, notably Australia, New Zealand, the United States and the European Union. The last coup in 2006 also unsettled relations in Fiji’s more immediate region, especially among some of its smaller Polynesian neighbours, contributing to Fiji’s unprecedented suspension from the Pacific Islands Forum. This article reviews Fiji’s foreign relations from the time of independence in 1970 through the period of successive coups to the 2014 elections. It also examines Bainimarama’s strategies in extending foreign relations in the broader international sphere as well as issues surrounding the ‘normalisation’ of relations with its traditional partners. 相似文献

AbstractInternational organisations active in Africa are often criticised for their ineffectiveness. So too is the European Union (EU), which is also accused of failing to assume a more prominent conflict management role in war-torn countries. This article examines the EU’s capacity and readiness to take on such a role in one such country, the former Republic of Sudan, home to Africa’s longest-running civil wars and the first ‘genocide’ of the 21st century. It begins by outlining the EU’s record in Darfur and the North–South Peace Process. Drawing upon 25 interviews and Hill’s ‘capabilities–expectations model’, it then questions whether the EU’s ‘capabilities’ (resources, instruments, unity) were ‘fit for purpose’ in Sudan’s hostile target setting. It concludes by identifying settings that have been more propitious for a conflict-related management function and by suggesting that the EU should better manage expectations about future security roles. 相似文献
