Asia 1型口蹄疫病毒多抗原表位真核表达质粒的构建及表达

为了构建Asia 1型口蹄疫病毒多抗原表位真核表达质粒,采用多表位基因串联策略,合成包括口蹄疫病毒T细胞和B细胞表位基因的基因片段F,该基因片段由结构蛋白VP1上的2个B细胞表位(VP1135-159aa、VP1194-211aa)基因、非结构蛋白3A和3D上的T细胞表位(3A21-35aa、3D795-803aa)基因组成。利用限制性内切酶HindⅢ、XbaⅠ将基因片段F克隆至真核表达载体pcDNA3.1(+)。经酶切鉴定、序列分析正确后,利用LipofectamineTM2000将阳性重组质粒pc-F转染至BHK-21细胞。Western-blot和IFA分析结果显示,所表达的蛋白大小约25.4ku,能够与Asia 1型口蹄疫病毒标准兔抗血清发生特异性反应,证实所表达的蛋白具有较好的反应原性。结果表明,Asia 1型口蹄疫病毒多抗原表位真核表达质粒pc-F构建成功,且能在BHK-21细胞中正确表达。     

口蹄疫病毒3D基因的真核表达及其表达产物的功能分析

利用RT-PCR技术扩增口蹄疫病毒(FMDV)编码RNA依赖的RNA聚合酶的3D基因,经测序确认后将其克隆到真核表达质粒载体pcDNA3.1( )中,重组质粒pcDNA3.1( )-3D经HindⅢ、XbaⅠ双酶切后转染至BHK-21细胞,用纯化的目的蛋白进行Western-blotting鉴定,并预测该3D基因表达产物的三级结构。结果显示,获得分子质量约55 ku的单一3D基因表达产物,其三级结构较为保守。利用RNA细胞内复制体系和荧光定量RT-PCR技术,证明3D基因表达产物在细胞内可促进口蹄疫病毒基因组的复制。     

鸡IL-18基因重组真核表达载体的构建及其表达产物的生物学活性

从pMDChIL18克隆质粒扩增获得了ChIL18全基因片段,并将其重组到真核表达载体pcDNA3.1( )。经酶切、质粒PCR鉴定和基因测序,鸡IL18全基因被正确重组到pcDNA3.1( )真核表达质粒上;将重组真核表达质粒pcDNA3.1( )ChIL18转染COS7细胞,转染细胞中含ChIL18基因的mRNA。SDSPAGE分析表明,表达产物是与ChIL18相符的约23ku的蛋白条带。鸡淋巴细胞转化试验表明,表达产物对鸡淋巴细胞具有明显诱导转化作用。     

亚洲Ⅰ型口蹄疫病毒VP1基因真核表达载体的构建及其免疫活性的初步分析

提取亚洲Ⅰ型口蹄疫病毒总RNA,用特异性引物通过RTPCR反应获得了约750bp的核酸片段,将其与pGEMTEasy载体连接,并转化大肠埃希氏菌JM109。重组质粒经PCR、酶切鉴定及测序,证明所克隆的片段含有完整的VP1基因;将重组质粒与表达载体pcDNA3.1( )分别用BamHⅠ和EcoRⅠ双酶切后连接,构建成重组表达质粒,转化宿主菌DH5α,筛选阳性克隆。测序结果证明,目的基因正确插入到表达载体中,命名为pcDNAV;用同样方法将IL18基因插入pcDNAV中,命名为pcDNAVI。用pcDNAVI免疫豚鼠,2周后加强免疫1次,每周用ELISA检测豚鼠血清中的抗体水平。结果表明,pcDNAVI可引发机体产生体液免疫。     

流行性乙型脑炎病毒E蛋白抗原表位区与猪细小病毒VP2真核表达质粒的构建及免疫效果

筛选了3个位于流行性乙型脑炎病毒(JEV)E蛋白上不同位置的抗原表位区基因,利用重叠PCR方法分别与猪细小病毒(PPV)VP2基因相连。将目的基因定向插入真核表达载体pcDNA3.1(+),构建了重组质粒pcDNA3.1-EA-VP2、pcDNA3.1-EB-VP2、pcDNA3.1-EC-VP2。将重组质粒在无免疫佐剂参与的情况下免疫BALB/c小鼠,同时设空质粒接种组及PBS缓冲液接种组作为空白对照,采用ELISA法、淋巴细胞增殖试验和流式细胞仪技术分别对抗体水平、脾淋巴细胞转化功能和小鼠外周血中CD3+、CD4+和CD8+T淋巴细胞数进行检测。结果显示,重组质粒pcDNA3.1-EA-VP2、pcDNA3.1-EB-VP2、pcDNA3.1-EC-VP2接种小鼠后,都能诱导小鼠产生JEV、PPV特异性抗体,并且促进脾淋巴细胞增殖。试验组与对照组比较差异极显著(P0.01);试验组小鼠CD3+、CD4+T淋巴细胞数在首疫后第7天高于对照组(P0.01),CD8+T淋巴细胞数在首免后第14天高于对照组(P0.01)。pcDNA3.1-EB-VP2免疫组小鼠的体液免疫和细胞免疫应答水平均高于pcDNA3.1-EA-VP2和pcDNA3.1-EC-VP2免疫组。表明,用JEV E蛋白抗原表位区基因与PPV VP2基因构建的真核表达质粒能够诱导小鼠产生良好的细胞免疫和体液免疫应答。     

猪传染性胃肠炎病毒核酸免疫昆明鼠的抗体应答

以猪传染性胃肠炎病毒 (TGEV)TH98株N基因的重组质粒 pHN为模板 ,Li 15和Li 2为上、下游引物扩增出TGEVTH98/N基因。将N基因与真核表达载体pcDNA3.1(+)分别用BamHⅠ、XhoⅠ双酶切后连接 ,构建了重组真核表达载体 pcDNA N。将昆明鼠随机分为 2组 ,分别肌肉注射 pcDNA N和pcDNA3.1(+) ,每只鼠 10 0 μg ,共免疫 3次 ,每次间隔 2周。首次免疫后第 0、14、2 1、2 8、35、39、4 7和 5 4d采血分离血清 ,采用ELISA检测抗体动态变化。注射pcDNA N组小鼠在首次免疫后第 39d检测到针对TGEVN蛋白的阳性抗体。     

口蹄疫病毒全长cDNA克隆的快速构建

利用RT PCR和 3′cDNA末端快速扩增 (RACE)技术分别扩增出包含口蹄疫病毒(FMDV)全基因组的 2个重叠cDNA片段 ,将其分别插入到载体 pcDNA3.1zeo( )和pGEM T Easy中 ,然后利用片段本身单一酶切位点将FMDV全长cDNA克隆至 pcDNA3.1zeo( )载体 ,经PCR扩增、酶切鉴定和序列测定。结果表明 ,重组质粒 (pcDNA3.1/FMDV)携带了FMDVOH/99基因组全长cDNA序列     

野鸭源新城疫病毒V蛋白基因在DF-1细胞中的表达及V蛋白的亚细胞定位

为研究野鸭新城疫病毒V蛋白基因在真核细胞中的表达及V蛋白的亚细胞定位,将新城疫病毒V基因片段与pcDNA3.1载体连接,构建pcDNA3.1-V重组表达质粒,经脂质体转染DF-1细胞后,用Western-blot验证该基因在DF-1细胞中的表达;用激光共聚焦显微成像技术确定V蛋白在真核细胞中的分布及亚细胞定位。结果显示,重组质粒pcDNA3.1-V在外源真核细胞DF-1中获得了表达,表达蛋白主要聚集于细胞质。研究结果为进行野鸭源新城疫病毒V蛋白的功能研究奠定了基础。     

Ea3-1E与mChIL-15融合基因的构建及其真核表达质粒的免疫效果

应用重叠扩增PCR技术将鸡堆形艾美球虫子孢子表面抗原3-1E基因片段(Ea3-1E)与编码鸡白细胞介素15成熟蛋白基因片段(mChIL-15)进行串联连接,并在这2个基因片段之间引入4个柔性连接肽SPGS,获得融合基因Ea3-1E-linker-mChIL-15。以pcDNA3.1(+)为载体构建并鉴定真核表达质粒pcD-NA3.1-Ea3-1E-linker-mChIL-15。应用磷酸钙法将质粒体外转染293T细胞,通过间接免疫荧光技术和免疫组织化学技术对重组质粒的体外瞬时表达进行检测。将重组质粒pcDNA3.1-Ea3-1E-linker-mChIL-15、pcDNA3.1-Ea3-1E和pcDNA3.1分别于14和21日龄经腿部肌肉注射免疫SPF雏鸡,28日龄时,除非免疫非感染组外,各组感染5×104个堆形艾美球虫孢子化卵囊,观察真核表达质粒的免疫保护效果。结果显示,成功构建了融合基因3-1E-linker-mChIL-15,转染后第30小时可检测到融合基因编码蛋白在293T细胞中的瞬时表达。与质粒pcDNA3.1-Ea3-1E相比,质粒pcDNA3.1-Ea3-1E-linker-mChIL-15二次免疫后可提供明显的抗球虫免疫保护效果,提高相对增重率(96.48%),降低卵囊排出率(68.35%),降低十二指肠病变记分等。抗球虫指数为183.78。     

弓形虫ME49虫株α-微管蛋白对神经瘤母细胞自噬的影响

为研究刚地弓形虫α-微管蛋白(TgTUBA1)与神经瘤母细胞(N2a)自噬的关系,利用TgTUBA1基因的CDS序列设计引物并引入HA标签序列,提取弓形虫ME49虫株总RNA并反转录获得cDNA,以cDNA为模板进行PCR扩增,获得目的片段,并克隆至pcDNA3.1(+)载体上。将构建的真核表达质粒pcDNA3.1(+)-HA-TUBA1转染至N2a细胞内,通过透射电子显微镜和荧光显微镜观察自噬小体,并通过Western-blot检测LC3Ⅰ/Ⅱ的表达变化。结果显示,成功构建了pcDNA3.1(+)-HA-TUBA1真核表达载体,Western-blot试验检测到50 ku大小的特异目的条带。与空载体组相比,pcDNA3.1(+)-HA-TUBA1转染N2a细胞48 h后,N2a细胞皱缩、突触减少;观察到特征性的自噬小体;自噬相关蛋白LC3Ⅰ/Ⅱ表达上调。本研究证明了TgTUBA1蛋白可以促进N2a细胞自噬现象的发生,这为宿主细胞抗弓形虫感染提供了新的研究思路。     

The Development of EU-Russian Relations

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. But the two sides would refrain from extreme measures and prevent their relations from sliding into real cold war.     

Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.     

The Balance Between Coercion and Accommodation

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.
