堆形艾美球虫广东株Ea1A基因的扩增与序列测定

根据GenBank上登录的堆形艾美球虫Ea1A基因序列 ,设计了 1对引物 ,以抽提的广东株的总RNA为模板 ,利用反转录 聚合酶链反应 (RT PCR)扩增获得了Ea1A基因部分片段 ,并将此片段克隆至pGEM T载体中 ,经PCR、限制性内切酶分析和克隆片段序列测定、比较 ,证实了克隆片段的可靠性     

金黄色葡萄球菌纤连蛋白结合蛋白A基因的克隆及其原核表达

根据GenBank中纤连蛋白结合蛋白A基因(fnbA)序列设计了1对特异性引物,以金黄色葡萄球菌基因组DNA为模板,进行PCR扩增;结果获得了3 600bp的DNA片段。将PCR产物克隆至pGEM T easy载体中,成功地构建了克隆质粒pGEM-fnbA。以HindⅢ和XhoⅠ双酶切pGEM-fnbA和pET28a( ),将纯化的基因fnbA亚克隆至pET28a( )中,构建了原核表达质粒pET28a-fnbA,并将其转化至E.coliBL21(DE3)感受态细胞中,经1 mmol/L IPTG诱导和SDS-PAGE分析,在约165 ku处出现了与预期目的蛋白一致的外源蛋白带。Western-blotting分析表明,该蛋白具有金黄色葡萄球菌的抗原性。     

环形泰勒虫裂殖体表面蛋白基因高免疫原性区片段的克隆及其表达

根据环形泰勒虫TaA12 72 1株裂殖体表面抗原 (TaSP)基因的序列设计了 1对引物 ,用PCR扩增出该基因长为 393bp的高免疫原性区片段 (SBxp)。将扩增产物连接到 pGEM TEasy载体上 ,转化入大肠埃希氏菌JM 10 9中进行克隆。随后将重组质粒 pGEM SBxp和表达载体 pGEX 4T 1分别以BamHⅠ、XhoⅠ酶切后 ,将酶切的目的片段SBxp亚克隆到原核表达载体中构建了重组表达载体 pGEX 4T 1 SBxp ,并将其转化到BL2 1宿主菌中。提取重组质粒经BamHⅠ、XhoⅠ酶切、PCR鉴定和测序确证后 ,阳性克隆用IPTG诱导表达。收集不同时间的菌液进行SDS PAGE电泳和Westernblotting检测。结果 ,SBxp基因在大肠埃希氏菌中成功表达 ,其表达产物为分子质量 4 3ku的融合蛋白 ,该产物能被牛环形泰勒虫阳性血清所识别。     

口蹄疫病毒多基因腺病毒穿梭质粒的构建

采用分步扩增连接的方法将口蹄疫病毒结构蛋白基因P1 2A、3C蛋白酶基因和 3D聚合酶基因按正确的读码框依次连接克隆入 pGEM T载体。然后 ,将切取的目的基因亚克隆入腺病毒穿梭质粒 pShuttle CMV。构建成功的 2个重组穿梭质粒经测序鉴定 ,含有完整的目的基因表达盒 ,穿梭质粒可与腺病毒骨架载体进行细菌内同源重组 ,以产生重组腺病毒载体     

口蹄疫病毒全长cDNA克隆的快速构建

利用RT PCR和 3′cDNA末端快速扩增 (RACE)技术分别扩增出包含口蹄疫病毒(FMDV)全基因组的 2个重叠cDNA片段 ,将其分别插入到载体 pcDNA3.1zeo( )和pGEM T Easy中 ,然后利用片段本身单一酶切位点将FMDV全长cDNA克隆至 pcDNA3.1zeo( )载体 ,经PCR扩增、酶切鉴定和序列测定。结果表明 ,重组质粒 (pcDNA3.1/FMDV)携带了FMDVOH/99基因组全长cDNA序列     

phyA基因的克隆及其新型表达载体的构建

直接以黑曲霉菌株F2 4 6基因组为模板 ,对植酸酶基因phyA的成熟肽 (分子长度为1347bp)进行了PCR扩增 ,再将PCR产物克隆入 pMD 18 T质粒 ,经DNA测序鉴定正确后 ,亚克隆入表达载体 pET30a 和 pMG36e ,成功构建了phyA基因 2种新型表达载体。植酸酶 phyA基因2种新型表达载体的构建 ,为进一步获得大量、高活性植酸酶以及研究和开发新型微生态制剂打下了基础。     

伪狂犬病病毒Min-A株gE基因的克隆及序列分析

参考GenBank中伪狂犬病病毒 (PRV) gE基因的序列设计了 1对引物 ,对PRVMin A株进行了PCR扩增 ,扩增产物克隆于 pGEM TEasy载体。对重组质粒进行限制性内切酶分析和基因测序 ,证实了克隆片段的可靠性。测序结果表明 ,目的片段包含 1个 174 0bp的开放性阅读框(ORF) ,编码由 5 79个氨基酸组成的多肽。同源性分析表明 ,PRVMin A株与PRVEa株、SH株gE基因的核苷酸同源性分别为 99.2 %、98.7% ;推导氨基酸的同源性分别为 98.6 %、97.2 %。     

山羊痘弱毒疫苗株TK基因的克隆及序列分析

以提取的山羊痘弱毒疫苗株HLJ-V9 DNA为模板,设计特异性引物并进行PCR扩增,获得了2078 bp的DNA片段,并将PCR产物克隆至pGEM-T Easy载体。酶切鉴定、PCR鉴定和测序结果表明,成功获得了TK基因,获得的TK基因内存在一个KpnⅠ酶切位点和编码区的早期转录终止信号TTTTTN(T)T。核苷酸和氨基酸同源性分析表明,弱毒疫苗毒株HLJ-V9 TK基因与参考毒株的同源性在95.5%~100%之间,说明羊痘病毒TK基因具有高度的保守性。     

鹅Ⅰ型禽副粘病毒融合蛋白基因3′端cDNA片段的分子克隆

参考禽副粘病毒的融合蛋白基因 (F基固 )cDNA序列 ,设计并合成了 1对引物 ,以分离到对鹅有致病力的Ⅰ型禽副粘病毒GPMV/QY97 1株的核酸 (RNA)为模板 ,用逆转录聚合酶链反应 (RT PCR)对其融合蛋白基因 3′端进行扩增 ,获得了预计的 884bp左右的片段。将该片段克隆进 pGEM TEasy质粒载体 ,获得重组质粒T GPMVF3′ ,采用限制性内切酶和PCR方法对该重组质粒进行鉴定 ,证明所克隆的片段即为目的基因片段 ,这为进一步开展鹅Ⅰ型禽副粘病毒的分子生物学研究奠定了基础     

胸膜肺炎放线杆菌APXⅡA基因的克隆及序列测定

用PCR法从胸膜肺炎放线杆菌 (APP) 7型的DNA提取物中扩增出大小为 2 873bp的APXⅡA基因片段 ;将PCR产物重组到质粒载体 pMD 18 T中 ,并对重组质粒进行酶切分析及序列测定。结果表明 ,克隆片段为完整的目的基因 ,该片段的核苷酸序列与国外分离株M 30 6 0 2的同源性达 99.7%。     

The Development of EU-Russian Relations

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. But the two sides would refrain from extreme measures and prevent their relations from sliding into real cold war.     

Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.
