牛蓝舌病病毒VP7抗体胶体金试纸条检测方法的建立

为建立一种快速检测牛蓝舌病病毒抗体的免疫层析方法,本研究通过胶体金标记蓝舌病病毒VP7蛋白,以兔抗牛IgG作为检测线,抗蓝舌病毒VP7蛋白单克隆抗体作为质控线,采用间接法制备了胶体金免疫层析试纸条。结果显示,该胶体金试纸条在蓝舌病病毒抗体阳性血清稀释度为1∶64时仍可检出;该试纸条与牛结核病、牛布鲁氏菌病、牛病毒性腹泻、牛巴氏杆菌病、牛地方流行性白血病的阳性血清均无交叉反应;与国外商品化试剂盒相比较,该试纸条检测结果与ELISA检测试剂盒(IDEXX)的符合率为92.7%(51/55)。结果表明,本试验初步建立的牛蓝舌病病毒抗体的胶体金免疫层析检测方法具有较好的特异性和稳定性,整个检测过程可在10 min内完成,能够快速检测样品血清,适用于现场快速检测。     

A群猪轮状病毒胶体金免疫层析试纸条的研制

为建立一种快速、简便的猪轮状病毒检测方法,采用柠檬酸三钠还原法制备胶体金颗粒,标记纯化的抗猪轮状病毒VP6蛋白单克隆抗体,在硝酸纤维素膜上喷加纯化的抗猪轮状病毒VP6蛋白多克隆抗体和羊抗鼠IgG,制成检测猪轮状病毒抗原的胶体金免疫层析试纸条。结果表明,所制备的试纸条用于检测猪轮状病毒感染的MA104细胞培养物,在检测线处出现红色反应带,未感染病毒的细胞培养物在检测线处未出现反应条带,上述培养物在质控线处均出现红色反应条带;进一步试验表明,试纸条检出病毒培养液(TCID50是10-6.25/0.1 mL)的最低限为1∶32稀释;用不同批次的试纸条重复检测,结果无差异;该试纸条不与相关的腹泻病毒猪传染性胃肠炎病毒、流行性腹泻病毒发生反应,所制备的试纸条具有一定的敏感性和特异性,重复性良好。     

猪瘟病毒胶体金免疫层析检测方法的建立及应用

为建立一种猪瘟病毒的快速检测方法,采用柠檬酸三钠还原氯金酸制备胶体金,选择15 nm胶体金标记兔抗猪瘟病毒抗体,对胶体金标记抗体采用低温超速离心法进行纯化,组装免疫层析试纸条。用试纸条对猪瘟可疑病料进行检测,同时用直接荧光抗体试验和RT-PCR做平行试验。结果显示,用试纸条检测猪瘟病毒,10 min左右即可显示结果,试纸条、直接猪瘟荧光抗体试验和RT-PCR的阳性检出率依次为36.36%(4/11)、45.45%(5/11)和72.72%(8/11)。表明,用胶体金免疫层析试纸条检测猪瘟病毒操作简便,需时短,可以用于猪瘟的流行病学调查。     

应用降落PCR检测减蛋综合征病毒的研究

根据已发表的减蛋综合征病毒 (EDSV)核苷酸序列设计了 1对扩增长度为 1 .1kb左右的引物 ,采取降落PCR对EDSV gDNA进行扩增。结果 ,从 2份EDSV鸭胚尿囊液、3份人工感染鸡输卵管组织液的核酸抽提物中 ,均扩增出与设计大小完全一致的目的片段 ,而对照样品的扩增均为阴性 ,该方法最低可检测到 32pg的EDSV gDNA。表明该降落PCR用于检测EDSV特异性和敏感性均较强 ,可用于减蛋综合征的诊断     

狂犬病病毒抗体免疫金标检测试纸的制备及其应用

为了建立一种快速检测狂犬病病毒抗体水平的方法,用浓缩提纯的狂犬病病毒CVS11株作为胶体金标记抗原,以金黄色葡萄球菌A蛋白为检测线上的捕获抗原,制备狂犬病病毒抗体水平检测试纸条。应用该试纸条进行临床样品检测,结果表明,以稀释后的犬全血或血清作为检测样品,试纸条检测的灵敏度为0.002U/mL,特异性试验证明该试纸条与犬细小病毒、犬瘟热病毒、犬腺病毒2型犬科疾病阳性血清没有交叉反应,稳定性试验证明该试纸条在室温下保存的有效期为12个月,检测结果与快速荧光抑制试验的符合率为95.12%。证实该狂犬病病毒抗体免疫金标检测试纸可以方便、快速、灵敏、特异地检测狂犬病病毒的抗体水平。     

鹅细小病毒胶体金免疫层析检测方法的建立

为建立一种快速、简便地检测鹅细小病毒(GPV)的方法,采用柠檬酸三钠还原法制备胶体金,将其标记于抗GPV VP3蛋白单克隆抗体2D5上,在硝酸纤维素膜上喷涂抗GPV VP3蛋白单克隆抗体4A8和山羊抗小鼠IgG抗体作为检测线和质控线,制成检测GPV的胶体金免疫层析试纸条。结果显示,该试纸条检测GPV的最低TCID50为1×104.15/mL,用制备的试纸条仅检测GPV为阳性,而检测其他主要水禽病的病原结果均为阴性;同一批次和不同批次的试纸条对GPV阳性样品的检测结果均无差异。使用该试纸条对已知的80份粪便样品进行检测,阳性及阴性样品的检测符合率分别为96.49%和100%。结果表明,制备的试纸条具有良好的特异性、敏感性和重复性,为"小鹅瘟"的快速诊断提供了新的方法。     

嗜水气单胞菌胶体金快速检测试纸条的研制

用柠檬酸三钠还原氯金酸制备胶体金颗粒,标记抗嗜水气单胞菌单克隆抗体并制备金标垫。将吸收垫、喷涂有抗嗜水气单胞菌单克隆抗体和羊抗鼠抗体的硝酸纤维素膜、金标垫及样品垫组装成免疫层析试纸条,建立嗜水气单胞菌的胶体金免疫层析快速检测方法。用灭活细菌与血清混合的模拟检测样本测定试纸条的特异性、灵敏度,结果显示:试纸条对豚鼠气单胞菌、温和气单胞菌、鳗弧菌、溶藻弧菌、荧光假单胞菌、副溶血弧菌等13种常见病原菌没有交叉反应,与嗜水气单胞菌有特异性反应,检测灵敏度为1×105CFU/mL,检测时间小于5min。所制备的嗜水气单胞菌胶体金免疫层析检测试纸条具有快速、简便、特异性高、适用于基层临床生产推广应用等优点。     

基于牛源布氏杆菌重组膜蛋白的胶体金免疫层析试纸条的研制

为研制一种快速检测牛源布氏杆菌的免疫层析技术,本研究对牛布氏杆菌外膜蛋白omp22进行表达纯化,并用新西兰大白兔制备omp22多克隆抗体,将omp22蛋白作为胶体金标记物,家兔抗牛Ig G用于检测线,多抗用于质控线,制备了胶体金免疫层析检测抗体试纸条。结果显示,成功诱导表达大小为74 ku的可溶性重组蛋白omp22,且纯化后的重组蛋白omp22的浓度为2 mg/m L,纯度在85%以上;该胶体金试纸条在阳性血清稀释至1∶64时仍可见清晰条带;与牛结核、牛蓝舌病、牛病毒性腹泻、牛口蹄疫、牛巴氏杆菌病、牛地方流行性白血病阳性血清均无交叉反应;与国外商品化试剂盒比较,试纸条检测结果与ELISA检测试剂盒(IDEXX)的符合率为94.2%(49/52),与牛羊布病抗体检测卡(KERNEL)的符合率为96%(49/50)。整个检测过程可在10 min内完成,肉眼即可判断。以上结果表明,该试纸条具有良好的重复性、敏感性和特异性,可适用于牛源血清中布氏杆菌病抗体的现场快速检测和筛选工作。     

二温式多重PCR同时检测鸡的6种呼吸道病病原方法建立

设计了6对分别针对鸡传染性支气管炎病毒(IBV)、禽流感病毒(AIV)、鸡毒霉形体(MG)、鸡传染性喉气管炎病毒(ILTV)、新城疫病毒(NDV)和滑液霉形体(MS)的特异性引物,根据反转录聚合酶链反应(RT-PCR)原理和多重PCR原则,优化建立了一次PCR反应同时检测6种禽呼吸道病原的二温式多重PCR.该方法可同时扩增出6条区段大小与设计相符的特异性片段,即IBV 1720 bp、AIV 1050 bp、MG 732 bp、ILTV 647 bp、NDV 310 bp、MS 207 bp,对人工感染鸡临床样品检测结果证明,该方法可用于临床诊断.

设计了 6对分别针对鸡传染性支气管炎病毒 (IBV )、禽流感病毒 (AIV)、鸡毒霉形体 (MG )、鸡传染性喉气管炎病毒 (ILTV )、新城疫病毒 (NDV)和滑液霉形体 (MS)的特异性引物 ,根据反转录聚合酶链反应 (RT PCR)原理和多重PCR原则 ,优化建立了一次PCR反应同时检测 6种禽呼吸道病原的二温式多重PCR。该方法可同时扩增出 6条区段大小与设计相符的特异性片段 ,即IBV 172 0bp、AIV 10 5 0bp、MG 73 2bp、ILTV 64 7bp、NDV 3 10bp、MS 2 0 7bp ,对人工感染鸡临床样品检测结果证明 ,该方法可用于临床诊断。     

The Development of EU-Russian Relations

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. But the two sides would refrain from extreme measures and prevent their relations from sliding into real cold war.     

Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.     

The Balance Between Coercion and Accommodation

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.     

U.S. and East Asian Integration

the U.S. is directly impacting on the future of the East Asian community. Therefore, finding ways to get along with the U.S. is crucial to the speed, direction, configuration and character of the East Asian community. In this paper, the author has analyzed the interests of the United States in East Asia and its stands towards the East Asian integration. The author concludes that it is to the interest of the United States to make more efforts to further join in the East Asian integration. And East Asia should accept and welcome the American participation.
