具有中和活性的鸭肠炎病毒单克隆抗体的制备与鉴定

为制备抗鸭肠炎病毒(DEV)的单克隆抗体(MAb)并对其生物学活性进行鉴定,本研究采用蔗糖密度梯度离心法提纯DEV鸭胚成纤维细胞适应毒,以此作为抗原免疫BALB/c小鼠,随后取小鼠脾细胞与SP2/0骨髓瘤细胞融合,有限稀释法克隆杂交瘤细胞株,经间接ELISA筛选,获得了1株稳定分泌抗DEV MAb的杂交瘤细胞株2E3。鉴定结果显示,该MAb为Ig M亚类,ELISA效价可达到1∶10~5以上;间接免疫荧光试验(IFA)中MAb可与细胞中增殖的DEV结合,Western-blot试验中MAb不与变性病毒蛋白作用,表明该MAb可能是构象表位的单抗;病毒中和试验表明,该MAb具有较强的病毒中和活性。上述结果表明,本试验获得的一株具有DEV中和活性的MAb杂交瘤细胞株为建立DEV的检测方法和DEV的相关研究提供了材料支持。     

分泌抗鸭瘟病毒单克隆抗体杂交瘤细胞株的建立

通过二次感染鸭瘟病毒的小鼠脾细胞与骨髓瘤细胞系SP_2/0融合试验和多次筛选及克隆化,建立了二株分泌抗鸭瘟病毒单克隆抗体的杂交瘤细胞株,用间接血凝和ELISA等方法证实二株细胞分泌的单克隆抗体对鸭瘟病毒具有良好的专一性,与同属病毒马立克氏病毒以及不同属的鸭肝炎病毒、鸡新城疫病毒无交叉反应,免疫球蛋白亚类鉴定,表示二株均为IgG_1。腹水效价,间接血凝达1280,ELISA达20000。这二株细胞已在体外培养下稳定地分泌特异性抗体达8个月之久。     

猪传染性胃肠炎病毒N蛋白单克隆抗体的制备及鉴定

以纯化的猪传染性胃肠炎病毒(TGEV)重组N蛋白免疫BALB/c小鼠,按照常规方法融合、筛选,最终获得2株抗TGEV N蛋白的杂交瘤细胞系,分别命名为2G7和2C2。2株单抗亚类均为Ig G1,腹水的间接ELISA效价均为1∶108。2株杂交瘤细胞连续培养20代,各代次的上清效价保持不变,均为1∶12 800,说明2株细胞分泌抗体的能力稳定。通过间接免疫荧光发现,制备的2株单克隆抗体能与感染TGEV的细胞发生特异性反应,在胞浆和细胞膜上有亮绿色荧光。结果表明,所制备的2株TGEV N蛋白的特异性单克隆抗体细胞株遗传稳定,分泌的抗体敏感性高,为TGEV病原检测奠定了基础。     

牛IFN-γ单克隆抗体的制备及鉴定

为进一步研究牛IFN-γ(BoIFN-γ)单克隆抗体在牛免疫学以及牛疫病诊断中的应用,用原核表达的重组牛IFN-γ(rHis-BoIFN-γ)免疫6周龄雌性BALB/c小鼠,免疫3次后取其脾细胞和骨髓瘤细胞SP2/0进行融合。用建立的间接ELISA方法筛选阳性杂交瘤细胞,经3～5次亚克隆,最终获得12株能稳定分泌抗体的阳性杂交瘤细胞株。Ig亚类鉴定结果显示这12株杂交瘤细胞均为IgG,其中5株为IgG1,3株为IgG2a,4株为IgG2b。这12株杂交瘤细胞在体外具有稳定分泌抗牛IFN-γ单克隆抗体的能力。Western-blot及间接ELISA证明这12株单克隆抗体均可特异性地结合rBoIFN-γ蛋白。间接免疫荧光试验显示,这12株单抗均与真核细胞BHK-21表达的rBoIFN-γ产生荧光反应,证明这些单抗能够与接近天然的BoIFN-γ反应,证实了单抗的特异性。ELISA叠加试验表明,3C2与1H4、1G6与1G4针对不同表位,1A10、1B2、2B9、3C4四株和1C1与1G6、1G4与3C2、1A10与1G8针对相同或相近的表位。     

猪链球菌2型人源分离株MRP单克隆抗体的制备与鉴定

为建立一种针对猪链球菌2型(SS2)毒力因子的特异性检测方法,研制了抗SS2溶菌酶释放蛋白(MRP)单克隆抗体(McAb),并对其生物学活性进行了分析.将表达、纯化的SS2菌株MRP免疫BALB/c小鼠,采用淋巴细胞杂交瘤技术将免疫鼠脾细胞与SP2/0骨髓瘤细胞融合,以纯化的MRP蛋白作为包被抗原,通过间接ELISA方法筛选获得了6株能稳定分泌抗MRP McAb的杂交瘤细胞系288、3G5、1G1、3G9、186和2C3.经用间接ELISA方法测定,单抗腹水的抗体效价均超过1105,杂交瘤细胞培养上清液的效价为1128～11 024.western-blotting鉴定结果表明,6株单抗均具有特异的生物学活性;以2B8单抗腹水和SS2多克隆抗血清建立的夹心ELISA方法可特异地检测MRP.     

猪传染性胃肠炎病毒重组N蛋白单克隆抗体的制备及鉴定

以纯化的猪传染性胃肠炎病毒(TGEV)重组N蛋白免疫BALB/c小鼠,取免疫小鼠的脾细胞与SP2/0骨髓瘤细胞进行融合,通过间接ELISA法对融合细胞进行筛选,采用有限稀释法对阳性细胞进行4次亚克隆,获得了3株能稳定分泌N蛋白单克隆抗体的杂交瘤细胞,分别命名为JY(4)-1、JY(4)-2、JY(4)-3.分别采用体外培养法、诱导腹水法和实体瘤法制备单克隆抗体,并用间接ELISA法检测其效价,结果显示,这3株杂交瘤细胞的细胞培养上清、小鼠腹水和血清的单抗效价均分别为1:512、1:10~5、1:10~5,其中以诱导腹水法制备的单抗产量和效价最佳.间接免疫荧光试验结果显示,这3株单抗均能与TGEV感染的ST细胞产生特异性免疫荧光,而不与猪细小病毒、猪生殖与呼吸综合征病毒、猪轮状病毒等发生交叉反应;经鉴定,3株单抗的亚类均为IgG2a,这3株杂交瘤细胞具有良好的遗传稳定性.     

沙丁胺醇单克隆抗体的制备及其鉴定

将沙丁胺醇 (salbutamol)琥珀酰化后 ,应用混合酸酐法与牛血清白蛋白偶联 ,免疫BALB/c小鼠 ,采用杂交瘤技术获得了 1株能稳定分泌抗沙丁胺醇单克隆抗体的杂交瘤细胞株。该单抗为IgG1亚类。经间接ELISA方法测定 ,其细胞上清抗体效价为 1∶3 .2× 10 6,腹水抗体效价为 1∶6.4× 10 8。该抗体与克仑特罗交叉反应率为 5 0 .0 0 % ,与莱克多巴胺交叉反应率为 0 .3 0 %。体外传代培养和冻存复苏后抗体分泌稳定。     

抗禽脑脊髓炎病毒单克隆抗体的制备及鉴定

利用提纯的禽脑脊髓炎病毒(AEV)VR株CEF适应毒制备抗原,免疫BALB/c小鼠,应用杂交瘤技术以间接ELISA筛选获得1株分泌抗AEV(VR)单克隆抗体的杂交瘤细胞.对制备的细胞上清及腹水进行单克隆抗体特性鉴定,经染色体计数、免疫球蛋白类及亚类测定、特异性试验、稳定性试验、SDS-PAGE电泳,表明该杂交瘤细胞染色体数目介于90-110条,该单克隆抗体亚类为IgG2a,分子质量为142 ku,ELISA测定细胞上清效价为11×104,腹水效价为11×106,与其它病原无交叉反应,抗体分泌稳定.     

非肌肉肌球蛋白Ⅱ型重链B羧基端多肽单克隆抗体的制备及鉴定

为了得到能够特异性识别非肌肉肌球蛋白Ⅱ型重链B(NMHCⅡ-B)的单克隆抗体,根据Marc-145细胞上NMHCⅡ-B羧基端的蛋白质序列,合成多肽,免疫BALB/c小鼠,取其脾细胞与SP2/0骨髓瘤细胞融合,用ELISA方法筛选阳性克隆;制备腹水并纯化;用间接ELISA和间接免疫荧光试验检测纯化抗体的特异性。结果显示,获得2株能稳定传代并分泌特异性抗体的杂交瘤细胞系;单克隆抗体的特异性鉴定显示,2株细胞系所产生的抗体能够特异性地与NMHCⅡ-B羧基端蛋白反应而且能与Marc-145细胞产生特异性的结合。这2株单抗的制备及鉴定为研究易感细胞上NMHCⅡ-B与猪繁殖与呼吸综合征病毒感染的关系奠定了基础。     

禽脑脊髓炎病毒单克隆抗体的制备与初步应用

利用提纯的禽脑脊髓炎病毒(AEV)Van Roekel株作为免疫原,免疫6周龄BALB/c小鼠,采取其脾细胞与SP2/0骨髓瘤细胞融合,用间接ELISA法筛选,间接免疫荧光法(IFA)和免疫组织化学法鉴定,经3次亚克隆得到了稳定分泌抗AEV单克隆抗体的杂交瘤细胞株F11和G2,制备了腹水,并利用该单克隆抗体初步建立了Dot-ELISA、间接ELISA和IFA等特异性检测AEV抗原的方法。     

The Development of EU-Russian Relations

The EU＇s strategy for Russia since the end of the cold war has to a large extent failed, and the direct consequences are the currently deadlocked EU-Russia relationship. There are three reasons for this： the EU and Russia＇s respective strength and status relative to each other have been changing since the beginning of this century; the EU＇s strategy towards Russia has been more fragmenting in recently years; and the American factor. The EU-Russia relations could continue to be in poor shape in the years to come, and it is very likely sometimes even further worsening. But the two sides would refrain from extreme measures and prevent their relations from sliding into real cold war.     

Young American China Watchers＇ Views on China

In this paper, the author has made a thorough study over the new generation of American China Watchers. Most of the pages are devoted to a detailed comparison between the old generation and the young generation of American China Watchers regarding to their background, education, and views over China. At the end of the paper, the author also gives some policy suggestions for both the Chinese and U.S. government based on the findings in this study.     

The Balance Between Coercion and Accommodation

1. Introduction
On the morning of April 1st, 2001, a U.S. EP-3 surveillance aircraft was conducting an intelligence flight along the Chinese territorial border in the airspace south-east of China＇s Hainan Island. Two Chinese fighter jets intercepted the spy plane and followed its flight, as is considered ordinary international practice. For reasons unknown, the EP-3 airplane made a sudden maneuver and crashed into one of the Chinese planes. The Chinese plane dropped into the sea, the pilot missing. The U.S. spy plane was also severely damaged. It intruded into Chinese territorial space and landed on Hainan Island without official permission from the Chinese side. After landing, the spy plane and 24 crew on board were detained by the Chinese authorities. This air collision episode was the first of its kind in the China-U.S. relations and an urgent diplomatic crisis ensued.     

U.S. and East Asian Integration

the U.S. is directly impacting on the future of the East Asian community. Therefore, finding ways to get along with the U.S. is crucial to the speed, direction, configuration and character of the East Asian community. In this paper, the author has analyzed the interests of the United States in East Asia and its stands towards the East Asian integration. The author concludes that it is to the interest of the United States to make more efforts to further join in the East Asian integration. And East Asia should accept and welcome the American participation.
